摘要

•Optimize the secondary-electron imaging of a cryo-scanning electron microscope to enable cellular secondary-electron imaging (CSEI).•On-the-fly location during cryoFIB milling by CSEI.•Enable three-dimensional(3D) location by CSEI.•No fluorescence labels and no additional imaging devices are required for CSEI, just using the basic function of a cryoFIB instrument.
