·AIM:To investigate the correlation between fundus blood flow parameters and carotid artery ultrasound blood flow parameters in patients with hypertensive retinopathy(HRP). ·METHODS:A total of 50 patients(22 left eyes and 28 right eyes)with HRP admitted to our hospital from June 2021 to June 2023 were retrospectively included as the experimental group, and 50 healthy physical examination subjects(22 left eyes and 28 right eyes)during the same period were included as the healthy group.Pearson correlation was used to analyze the correlation between fundus blood flow parameters and carotid artery ultrasound blood flow parameters. ·RESULTS:The AUC values of fundus blood flow parameters and carotid artery ultrasound blood flow parameters and their combined diagnosis of HRP were 0.853,0.844 and 0.935,respectively.Pearson correlation analysis showed that carotid systolic peak blood flow velocity was negatively correlated with foveal avascular zone(FAZ)area,FAZ circumference and non-circularity index, and positively correlated with macular vascular density(all P<0.05).The end-diastolic blood flow velocity was positively correlated with FAZ area and macular vascular density(all P<0.05).The internal carotid artery resistance index was positively correlated with FAZ area(P<0.05). ·CONCLUSION:The combination of fundus blood flow parameters and carotid artery ultrasound blood flow parameters in the diagnosis of HRP has good application value in the diagnosis of HRP.
Objective To explore the effects and mechanisms of bone marrow mesenchymal stem cell(BMMSC)-derived exosomes(BMMSC-Exo)on hypoxia/reoxygenation(H/R)-induced injury in rat cardiomyocyte cell line(H9c2).Methods BMMSC-Exosomes were isolated by ultracentrifugation. The cells were divided into three groups:control,H/R,and H/R+BMMSC-Exo(H/R+Exo).A hypoxia/reoxygenation(H/R)injury model was es-tablished by exposing cells to 12 hours of hypoxia followed by 6 hours of reoxygenation.Flow cytometry was used to detect cell apoptosis,DHE staining was used to assess cellular ROS levels,JC-1 immunofluorescence staining was used to evaluate mitochondrial membrane potential, and Western blot was used to detect mitochondrial autophagy-re-lated proteins. Results BMMSC-Exo treatment significantly alleviated oxidative stress, restored mitochondrial mem-brane potential, reduced mitochondrial autophagy levels, and effectively decreased cardiomyocyte apoptosis.Conclu-sions Bone marrow mesenchymal stem cell-derived exosomes alleviate H/R-induced cardiomyocyte injury.
Objective To investigate the expression levels of platelet microparticles(PMPs)in patients with diabetic nephropathy(DN)and their relationship with renal injury. Methods Thirty DN patients,30 T2DM patients without DN and 30 healthy controls were enrolled.Flow cytometry was used to detect the quantity and phenotype of PMPs,and ELISA was used to measure the levels of plasma renin, angiotensin Ⅱ(angiotoninⅡ,AngⅡ),vascular endothelial growth factor(vascular endothelial growth factor,VEGF)and transforming growth factor-β1(transforming growth factor-β1,TGF-β1).Kidney function was determined by blood biochemistry. Results The quantity of PMPs in the DN group was(1 564±346)particles/μL,which was significantly higher than that in the non-DN group(1 246±312)particles/μL and the control group(1 223±299)particles/μL(P<0.05).The PMPs in the DN group mainly expressed CD62P and CD41a,accounting for(76.5±12.3)%.Moreover,the levels and activation of PMPs increased with the progression of DN.The quantity of PMPs was positively correlated with renin and other factors(r=0.56-0.62,P<0.05),negatively correlated with the glo-merular filtration rate(GFR)(r=-0.64,P<0.05),and positively correlated with the urinary albumin excretion rate(UAE)(r=0.66,P<0.05).PMPs were also an independent influencing factor of UAE(β=12.34,P<0.05).Conclusion PMPs expression is elevated in DN patients, which may be associated with renal injury, but it is insufficient to confirm its role in the pathogenesis of DN.
Objective To explore the effect of cancer related anemia on the abundance of SARS-CoV-2 specific memory CD8+ T cells in peripheral blood. Methods A total of 34 HLA-A*11∶01 positive patients with middle- and late-stage tumors admitted in the Rocket Force Medical Center of PLA from June to July 2023 were enrolled in this study. According to hemoglobin (HGB) level, they were divided into non anemia group (≥120 g/L, n=14), mild anemia group (90~120 g/L, n=14), and moderate to severe anemia group (< 90 g/L, n=6). After their peripheral blood mononuclear cells were isolated, memory CD8+ T cells were stimulated by the epitopes of SARS-CoV-2 and then expanded. Positive epitopes were screened using enzyme-linked immunosorbent spot (ELISPOT) assay, and then MHC-peptide Tetramers were constructed. The proportion and number of Tetramer+ cells and the number of ELISPOT spots were detected in the expanded cells to reflect the abundance of SARS-CoV-2 specific memory CD8+ T cells. The relevance of patients' HGB level with the proportion and number of Tetramer+ cells and the number of ELISPOT spots was analyzed. The differences of the proportion and number of Tetramer+ cells and the number of ELISPOT spots was analyzed in different groups. Results Among the 34 patients, there were 25 males and 9 females, at an age range of 36~78 (median 57) years. The proportion and number of Tetramer+ cells and the number of ELISPOT spots were positively correlated with HGB level (P<0.05). The proportion and number of Tetramer+ cells and the number of ELISPOT spots were significantly decreased in patients with moderate to severe anemia than the non anemia patients (P<0.05). Conclusion The abundance of SARS-CoV-2 specific memory CD8+T cells in peripheral blood is positively correlated with patients' HGB level, indicating that CRA may increase the susceptibility of patients to viruses by reducing the abundance of memory CD8+T cells in peripheral blood.
