The primary focus of my laboratory's research is the regulation of pulmonary vascular tone and structure and the pathogenesis of pulmonary arterial hypertension, with an emphasis on the interactive roles of vasoactive factors such as nitric oxide, endothelin-1, thromboxane, serotonin, and sphingosine-1-phosphate. Various experimental preparations, including catheterized rats and mice, perfused rat and mouse lungs, isolated pulmonary artery rings, and cultured vascular smooth muscle and endothelial cells, are used in physiological, pharmacological, and molecular biological experiments to investigate mechanisms of control of vascular tone and structure in both normotensive and hypertensive lungs. Current focus is on the role of Rho GTPases, such as RhoA and Rac1, and their downstream effectors in mediating the cellular effects of vasoactive factors. Students and trainees play a direct role in this research by assuming primary responsibility for the design, performance, analysis, presentation, and publication of projects related to the overall goals.
