My research team has made seminal contributions to the discipline of plant pathology in particular molecular plant-pathogen-microbe interactions. We were among the first to prove that resistance induced in plants towards fungal pathogens is based on recognition of fungal effectors, which is subsequently followed by a very efficient defence response, the hypersensitive response (HR), a particular type of programmed cell death. In these studies we have used the Cladosporium fulvum-tomato pathosystem as a model. We have characterized numerous effectors that play a role in virulence. In the presence of the cognate Cf resistance proteins they trigger Cf-mediated defence responses including the HR. We have identified four different mechanisms that enable the fungus to avoid recognition of its effectors by its host (point mutations, frame shifts, deletions, transposon insertions). We have characterized the virulence functions of two effectors, Avr2 and Avr4, in detail. Avr2 inhibits several cysteine proteases, including Rcr3. In the presence of the tomato Cf-2 resistance protein Avr2 triggers an HR that also requires Rcr3. Avr4 is a chitin-binding protein that protects the fungus against plant chitinases. In the presence of resistance protein Cf-4, Avr4 triggers the HR. Avr4 proteins encoded by virulent alleles in strains of C. fulvum are no longer recognized by Cf-4 plants, but still bind to chitin, suggesting that chitin-binding by Avr4 represents a basal protective virulence factor. We have also studied signal transduction responses triggered by effectors and mediated by cognate Cf proteins. Our findings have lead to several patents that are exploited in disease resistance breeding. Recently, we have discovered by genome sequencing that C. fulvum can be considered a type species of the Dothideomycetes, including Mycosphaerella fijiensis, M. graminicola, Cercospora nicotiana and C. beticola.
