One of his great achievements was to reveal the A-factor regulation cascade in the streptomycin producer Streptomyces griseus. First discovered by Dr. Khokhlov in Moscow in the late 1960s, factor A is a microbial hormone that triggers morphogenesis and secondary metabolism in S. cinerea. The Beppu professor rediscovered the A-factor in the early 1980s and began working on it with then assistant professor Dr. Horiuchi. The A-factor regulatory cascade appears in a long list of important papers in major journals authored by Dr. Horinouchi's group. When factor A is produced in a growth-dependent manner at critical concentrations, as low as 10-9 M in S. griseus, it binds to the factor A-specific receptor (ArpA), which binds to the adpA promoter and removes ArpA from the dissociation in the promoter. The transcriptional activator AdpA then activates the transcription of many genes required for secondary metabolism and morphological differentiation, forming the AdpA regulon, which includes strR, which encodes the streptomycin biosynthetic pathway-specific transcriptional activator.
