Evaluation of correlation between TPase/DPDase and capecitabine/5-fluorouracil

Objective To evaluate the correlation between biologic enzymes of thymidine phosphorylase(TPase) and dihydropyrimidine dehydrogenase(DPDase) and chemotherapeutic drug concentration of capecitabine(CAP) and 5-Fluorouracil(5-Fu) in breast cancer(Ca) and normal breast tissues(Nor).Methods In 26 cases of Ca and Nor tissues identified by pathology,TPase and DPDase protein levels were quantified by ELISA.The high-performance liquid chromatography(HPLC) was performed for detection of CAP and 5-Fu concentrations′ in these 26 individuals.Results The average TPase level in Ca was 386.1163 ng/(mg protein),and 42.5682 ng/(mg protein) in Nor.The average DPDase level in Ca was 209.528 ng /(mg protein),and 339.368 ng /(mg protein) in Nor.The range of CAP/5-Fu ratio was 0.0804-5.0000.Conclusions At the level of P=0.05,the TPase level was higher in Ca than in Nor,while there was no difference in the DPDase levels between Ca and Nor.A strong correlation between DPDase levels in Ca and Nor was observed(Spearman coefficient=0.43rs0.05/2(26)=0.390,P0.05).Ratio of TP/DPD is superior to efficacy of TPase and DPDase in predicting efficacy of CAP and 5-Fu.
