Influence of Mg2+ and vitamin E on TNF-α and caspase-3 contents in CCl4-induced liver injury

Objective To explore the influence of Mg~(2+) and vitamin E on TNF-αand caspase-3 contents in CCl_4-induced liver injury. Methods Fifty 3-month-old Kunming mice were randomized into model group,normal control group,Mg~(2+) group,vitamin E group; Mg~(2+) + vitamin E group.At day 11 of feeding,the mice in control group were intraperitoneally injected with soybean oil solution,while the mice in the other groups were intraperitoneally injected with soybean oil solution mixed with 0.15%CCl_4.Blood was separated from eyes to detect the level of serum ALT after treatment for 24 h.The mice were sacrificed to calculate hepatosomatic index,and the liver homogenate was prepared to determine GSH-Px,NOS activities and MDA,GSH,NO,TNF-α,caspase-3 contents.Results Compared with control group,hepatosomatic index,MDA content,,ALT and NOS activities increased in model group,and NO,TNF-α,caspase-3 contents also increased,while GSH content,GSH-Px activity decreased.Compared with model group,hepatosomatic index,the contents of MDA,ALT and NOS activities and the contents of NO,TNF-α,caspase-3 decreased in Mg~(2+) group and vitamin E group,while the content of GSH and GSH-Px activity increased.The effect in Mg~(2+) + vitamin E group was better than that in Mg~(2+) group and vitamin E group.Conclusion Mg~(2+) and vitamin E can antagonize and eliminate free radicals,increase the activity of antioxidant enzymes,inhibit the secretion of TNF-αand reduce caspase-3 content.Mg~(2+) combined with vitamin E has a synergistic protective effect on CCl_4-induced liver injury.
