Creation and Characterization of a Wheat/Rye Small Fragment Chromosome Translocation Line

A wheat/ rye small fragment chromosome transiocation line, WER-1-2, wasisolated from 8 pollen-derived plants of the F_1 hybrid of Chinese Spring and M27(1R/1D substitution line). Cytological observation including C-banding andfluorescence in situ hybridization (FISH) provided evidence that the original spike,from which line WER-1-2 was derived, contained one intact chromosome 1R andone abnormal 1R, of which the long ann end was deleted and translocated onto awheat chromosome. This translocation appeared to be the product of abnormal mitosisduring anther culture in vitro, not the product of aberrant meiosis. The resultsindicated that anther culture is an effect and expeditious way for transferring ofchromosome segments from distant species to wheat.
