•Rv1027c–Rv1028c encode functional KdpDE two component system in M. tuberculosis.•His642 and Asp52 are identified as phosphorylation sites.•Phosphorylation sensitive interaction of the KdpDE protein by SPR is reported.•Co-transcription of KdpD and KdpE genes observed under K+ ion limitation conditions.
