Fluorescence microscopy is a popular tool for imaging biological samples. The resolution of a microscope has been, until recently, limited by the diffraction limit of light, typically to ~250 nm in fluorescence microscopy. Localization-based super resolution microscopy has improved the final image resolution by roughly a factor of 10 and, in some cases, more. Here, we provide an overview of localization-based super resolution techniques and modifications for multicolor, three dimensional, and anisotropic imaging.
