We describe a method for the generation of antibodies against plant stress-related proteins in crude biochemical samples, such as proteins in suspension medium, by two-dimensional electrophoresis/phage panning (2D-PP). 2D-PP involves two-dimensional resolution of proteins, blotting of the proteins onto a nitrocellulose membrane, and screening of a phage antibody library. Phage antibodies were selected through 4 rounds of panning from the human synthetic single-chain Fv (scFv) library (Griffin.1). These antibodies are functional in Western blotting and are potential candidates as markers for the loss of regeneration capacity.
