•Enhanced activation of NMDA receptors in culture cause generation of new dendritic spines.•New spine formation involves entry of calcium ions through the calcium channel Orai1.•Spine formation in control neurons is reduced by dantrolene, a calcium store antagonist.•Caffeine releases calcium from stores, produces lower calcium rise in Orai1-deficient cells.•Calcium stores are important in formation of new dendritic spines.
