M2 polarization of macrophages by Oncostatin M in hypoxic tumor microenvironment is mediated by mTORC2 and promotes tumor growth and metastasis.

•Hx-Ca-CM secreted Oncostatin M regulates M2 polarization via mTORC2.•Hx-Ca-CM induced M2 polarization via Oncostatin M is affected by mTORC2 through Akt and not PKC-α.•In vivo myeloid specific rictor abrogation resulted in regression of 4T1 tumor.•It also exhibited diminished recruitment of macrophages and their M2 polarization.•It also impeded distant tissue metastasis to liver, lung and bone.
