236Background: Quantitative analysis of formalin-fixed paraffin-embedded (FFPE) prostate samples post radical-prostatectomy (RP) can help guide management of post-operative prostate cancer (PCa) patients. We have previously published on the results of our systems (precise) pathology platform which includes integration of clinical data with immunofluorescent (IF) morphology and biomarkers to predict disease progression in post-RP patients. We extended this approach by developing quantitative image features that further refine risk discrimination by directly competing with Gleason through the incorporation of biological attributes. Methods: Tissue microarrays representing a 892 RP population from Roswell Park Cancer Institute and the Henry Ford Hospital, with median 8 years follow-up were stained with a multiplex IF assay incorporating CK18 (epithelial cells), CK5/6 (basal cells), AMACR, the Androgen Receptor (AR), and Ki67.
