•Ubiquitin-specific protease 8 (USP8) regulates inflammation.•Cecal ligation and puncture (CLP) is a model of sepsis-associated encephalopathy.•We improved the CLP model and demonstrate a reduced mortality rate.•USP8 is protective against CLP-induced neuroinflammation.•USP8 is protective against CLP-induced cognitive and motor impairments.
