•Three peptidomimetics with 1, 2, and 3 Trp afford insights to interactions in complex proteins.•Molar absorptivity/quantum yield support presence of interactions between Trp units.•Quenching by acrylamide and triethylamine is dynamic in nature.•Quenching process is found to depend heavily on the solvent.•Quenching efficiencies/rates support presence of interactions involving Trp.
