•CysLT2R played positive role in LPS-induced microglia M1 polarization and negative role in IL-4-induced M2 polarization.•CysLT2R selective antagonist HAMI3379 could inhibit the activation of microglia M1 polarization and NF-κB pathway.•HAMI3379 promoted microglia polarization toward M2 phenotype which may exerts neuroprotective effects.
