•TaPUB26 overexpression enhanced ROS accumulation and affected Na+/K+ homeostasis.•TaPUB26 overexpression decreased the salt tolerance of Brachypodium distachyon.•High salinity and temperature, drought, and phytohormones upregulated TaPUB26.•TaPUB26 functions may be achieved via interactions with other proteins like TaRPT2a.•TaPUB26 was a negative regulator of salt-stress tolerance.
