Evaluation of nephroprotective effect of aqueous extract of terminalia chebula retz. fruits in gentamicin induced acute kidney injury in wistar albino rats

The aim of the study is to evaluate the nephroprotective effect of aqueous extract of Terminalia chebula Retz. fruits in gentamicin induced acute kidney injury in Wistar albino rats. 72 Wistar albino rats were randomly divided into 9 groups. Acute kidney injury (AKI) was produced with 80 mg/kg intraperitoneally (i.p.) injection of gentamicin for 7 days. α-lipoic acid 100 mg/kg i.p. served as the active control, while TC was given in two doses (200 mg/kg and 400 mg/kg orally) for 10 days as preventive and curative groups before and after the induction of AKI respectively. Distilled water, 1 ml/day orally for 10 days, served as the vehicle control. The preventive and curative effects of TC on nephrotoxicity were assessed by serological and histopathological parameters. Intraperitoneal administration of gentamicin produced renal dysfunction as evidenced by increased urine creatinine, serum creatinine, blood urea, serum potassium, MDA levels and decreased urine output, serum sodium, superoxide dismutase, glutathione levels and histopathological alteration of kidney architectures. Preventive and curative administration of 200 mg/kg and 400 mg/kg of T.C. resulted into a significant restoration of renal function markers, antioxidant parameters and histopathological architecture of kidney as compared to disease control group. Because of antioxidant properties, aqueous extract of Terminalia chebula Retz. fruits at 200 mg/kg and 400 mg/kg doses exerts nephroprotective effect and can be use to prevent & treat free radical mediated acute kidney injury.
