•We simulated dispersion of NH3 and CH4 dispersion from an idealized animal feedlot.•The NH3 deposition near the feedlot is inverted from changes to the NH3:CH4 ratio.•The inversion strategy is tested using the synthetic observations.•The sampling strategies are optimized using a sensitivity analysis.
