182 demonstrated 6 and 13 fold upregulation respectively in neonatal acid-treated rats (p<0.001vs controls).Expression profiles of miR-182-5p and miR-183-5p were validated using quantitative RT-PCR (fig.1).Bioinformatic analysis using miRTarBase database identified two transcription factors, FOXO1 and FOXO3a as validated targets for both miR -183-5p and miR-182-5p.In Western Blots, rCC extracts from neonatal acid-treated rats exhibited downregulation of FOXO1and FOXO3a expression compared to saline-treated controls, indicating miRNA-3'UTR target gene interaction between miR-182 cluster and FOXO1/ FOXO3a transcripts.Moreover, the experimental group also exhibited activation of AKT pathway by significant phosphorylation at Ser 474 compared to saline controls.CONCLU-SIONS: Early esophageal acid exposures result in long-term significant upregulation of miR-182 and miR-183 and activation of AKT pathway in rCC.These increases may be mechanistically involved in previously reported molecular alteration and synaptic plasticity in anterior cingulate cortex following early-life esophageal acid exposures.
