A role for phosphoinositide 3-kinase delta in neutrophil trafficking

We report here that PI3K catalytic subunit, p110delta is present in endothelial cells and it participates in neutrophil trafficking by modulating the proadhesive state of endothelial cells in response to TNFalpha. Administration of the PI3Kd selective inhibitor, IC87114, to animals reduced neutrophil tethering to and increased rolling velocities on TNFalpha-activated microvessels in a manner similar to that observed in p110d(-/-) mice. Inhibition of this isoform in endothelium reduced TNFalpha-induced Akt-phosphorylation and diminished neutrophil attachment in flow.
