•An NIR fluorescent probe (DDAP) was developed to sense Penicillin G acylase.•The endogenous PGA was characterized visually stained by DDAP on native PAGE-gel.•The hydrolysis of a series of β-lactams confirmed the hydrolase property of PGA.
