Expression of MCP-1 in white adipose tissues induce the resistin-hypersecretion in type 2 diabetes

Abstract Introduction Inflammation induced by MCP-1 plays an important role in the development of type 2 diabetes. Obese mice with MCP-1 deficiency showed an improvement of insulin resistance. However, type 2 diabetic model mice with MCP-1 expression have not been analyzed yet. To directly display the risk factor of MCP-1 in type 2 diabetic states, we have developed Irs-2-/- with MCP-1 expression (Irs-2−/−MCP-1Tg) mice. Methods Irs-2−/−MCP-1Tg mice were prepared by intercrossing with mating of female Irs-2--/+MCP-1Tg/+ × male Irs-2--/+MCP-1Tg/+ mice. After glucose tolerance test and insulin tolerance test, the liver, skeletal muscle, white adipose tissue, and pancreas were harvested and analyzed by Real time PCR, chemical analysis, and histologic analysis. Results The exacerbation of impaired glucose tolerance and insulin resistance in the Irs-2−/−MCP-1Tg mice was induced by resistin-hypersecretion and impairments of glucose metabolism. Conclusion MCP-1 expression might be useful as an indicator for exacerbation of type 2 diabetes status. Additionally, the resistin will be one of the important targets for the treatment of type 2 diabetes with inflammation.
