Sodium Ion Channels As Potential Therapeutic Targets For Cancer Metastasis

Many studies have shown the involvement of voltage-gated sodium channels (VGSCs) in cancer metastasis. However, a comprehensive review of using VGSCs as therapeutic targets for metastatic cancer drug development is still lacking. Here, we discuss the structural differences across VGSC subtypes and in the isoforms involving metastatic cancers. These structural variations are the basis for designing drug molecules with potential subtype selectivity. We also summarize the expression and activities of different VGSCs in certain types of cancer and possible mechanisms of action. Finally, we elaborate on the molecule-binding sites of VGSCs, emphasizing locations with potential in metastatic cancer drug development.
