•TSP-1 is highly expressed in the GCF of patients with chronic periodontitis.•TSP-1 is highly expressed in the inflammatory gingival tissues of rats.•Anti-TSP-1 inhibits the expression of MMP2, MMP9, RANKL/OPG in Pg•LPS-treated hPDLFs.•TSP-1 promots Pg•LPS-induced periodontal tissue destruction by the p38MAPK pathway.
