•Absolute MPTP dose differed with acute recovery versus severe acute neurotoxicity.•Body weight and mean brain volume did not differ across outcome groups.•Absolute, over weight-based, dose improves risk estimation of severe neurotoxicity.•Histopathology supports neurotoxicity at higher absolute MPTP doses.•Absolute MPTP dose should be used for intra-carotid MPTP in non-human primates.
