Validation Of New Hplc Method Determination Of Aflatoxin M1 In Raw Milk

The synthesis and full characterisation of the advanced materials, used for the treatment of infested natural products are based on the monitoring and control methodology and on the types of analyses for biologically infested samples. Usually, around 1-3% of aflatoxin B1 present in fodder are metabolised to aflatoxin M1 and concentrated in milk. Its presence with relative stability, as toxic compound, in raw and processed milk gives toxicity even with future processing - pasteurisation. The new HPLC method optimisation, in order to validate it, was performed in order to determine, by using an ordinary system (Dionex UltiMate3000 modular system having an UV detector), aflatoxin M1 metabolite in raw milk samples. The liquid liquid extraction process in the sample preparation stage was laborious but, at the end the recovery of the analyte was stable 34.67%. The quantification limit LLOQ obtained by chromatographic parameter optimisation, for aflatoxin M1 was 2.88 ng/g in the raw biological material, making this value lower than those accepted by the actual East-European normatives and regulations.
