•Isoliquiritigenin (ISL) treatment reduces blood glucose level and alleviates insulin resistance in high fat diet induced diabetic mice.•ISL treatment promotes hepatic glycogenesis and inhibits hepatic lipogenesis and fat accumulation.•ISL rapidly activates AMPK and inhibits mTORC1.•ISL reduces cellular ATP production.
