The effects of a chimeric adenoviruses(Ad)on the infection and growth inhibition of the human leukemia U937cells in vitro

Objective To investigate the effects of a chimeric adenoviruses(Ad)Ad5f11pTPEGFP on the infection and growth inhibition of the human leukemia U937cells.Methods The gene transduction efficiency and replication-competent ability were evaluated through inverted fluorescence microscope,fluorescence-activated cell sorting assay(FACS)and electron microscopy;Cellular proliferation was measured using the Cell Counting Kit-8(CCK-8);Cell apoptosis was analyzed by flow cytometry with AnnexinVAPC/7-AAD and the cleavage of enzyme PARP is detected by Western blot.Results The chimeric Ad Ad5f11pTPEGFP was significantly more effective than control Ad5GFP on infection of leukemia U937cells(P 0.05)and it could replicate within the chimeric Ad;Compare to Ad5GFP,the chimeric Ad caused more significant cytotoxicity of leukemia U937cells;The cell apoptosis rate of leukemia U937cells induced by Ad5f11pTPEGFP was significantly higer than that of Ad5GFP(P 0.05)and the level of cleavage of enzyme PARP increased with time.Conclusion The chimeric Ad Ad5f11pTPEGFP could infect leukemia U937cells effectively and replicate within them,also it can inhibit U937cells growth and induce cells apoptosis greatly.
