•Polyamine spermine can alleviate the clinical symptoms of EAE mice.•Spermine suppresses the activation of CD4+ T cells and polarization of Th1/Th17 cells by inhibiting ERK phosphorylation.•Our research disclosed the therapeutic potential of spermine and MAPK/ERK inhibitors in EAE/MS.
