Transcriptomic insights of Auxin Transport in Gymnema sylvestre R. Br

Gymnema sylvestre R. Br. is an important medicinal plant with antidiabetic properties. This is a non-model, slow growing medicinal plant and, except a transcriptome study, no genomic data is available in public domain. Understanding on cellular transport at transcriptome level was investigated for auxins being important plant hormone modulating many growth and developmental processes in plants. From a huge transcriptome date, we reported four genes Auxin Influx Carriers, Transport Inhibitors Response 1, Auxin Efflux Carriers and Auxin Response Factor. The sequences of these genes were used to prepare phylogeny of the nearest gene available in public domain and the model of the protein was predicted based on amino acid sequences. The analysis revealed that Auxin Influx Carriers in G. sylvestre (MT911901) is made up of 1461 bases and shares maximum similarity with Coffea eugenioides auxin transporter-like protein 2 (XM_027319767.1). Out of the five models generated through i-TASSER, the first model had C-score = −1.18, estimated TM-score = 0.57 ± 0.15 and the estimated RMSD = 10.0 ± 4.6 Å. The Transport Inhibitors Response 1 in G. sylvestre (MT911902) is made up of 1761 bases and is most similar to the Coffea eugenioides Transport Inhibitor Response 1 (XM_027304015.1). From this transcriptome analysis in G. sylvestre, three coding DNA sequences encoding PINs (two encoding PIN3; OP494270 and OP494271, one encoding PIN8; OP494272) and Auxin Response Factor (OP515523) are identified and submitted to the NCBI gene repository.
