•Posttransplant cyclophosphamide (PTCy) following hematopoietic stem cell transplantation (HSCT) contributed to impairing the graft-versus-leukemia effect and ameliorate graft-versus-host disease.•PTCy suppressed donor-derived alloreactive T cells except for CD44+ memory T cells.•The decreased T-cell chimerism with PTCy might associate impairing alloreactivity.
