•• Vitamin D restricts Zika virus infection and modulates the expression of a wide range of genes involved in the host's response to the virus.•• Vitamin D down-regulates the expression of cytokines and chemokines in Zika virus-infected macrophages.•• Vitamin D up-regulates the expression of antiviral response genes related to macrophage degranulation.•• Vitamin D up-regulated the expression of antimicrobial peptides, such as LL-37, which inhibits ZIKV infection and replication in macrophages.
