•SlPROPEP positively regulates tomato resistance by increasing the levels of reactive oxygen species and pathogenesis-related genes.•SlWRKY6 could directly bind to the promoter region of SlPROPEP and activate its transcription.•SlPep is involved in tomato-P. infestans interactions by modulating the JA/ET signaling pathway.
