•Casein kinase I family members play a crucial role in regulating autophagy, providing insights into the maintenance of cell homeostasis.•Silencing CK1δ/ε results in an increased autophagic flux, leading to suppressed proliferation and viability of cancer cells.•Inhibition of CK1δ/ε affects the process of tumorigenesis.•CK1δ/ε regulates ULK1-mediated autophagy in lung cancer and melanoma, suggesting its potential as a therapeutic target for these types of cancer.
