•Artemisia argyi essential oil was visualized by TLC-direct bioautography.•Rapid identification of bioactive compounds by 2D GC × GC-TOFMS.•In vitro antibacterial activity of six compounds were determined.•A rapid and efficient method for determination of antibacterial substances was established.
