Design, Synthesis and Biological Evaluation of Novel Pyrido-[2,3-d]-pyrimidin-2-amine Analogues as Antimycobacterial Agents

•Thirty-eight Pyrido[2,3-d]pyrimidin-2-amine analogs were synthesized, characterized, and tested against Mycobacterium tuberculosis (Mtb) H37Rv.•P35 showed significant activity with MIC 7.18 μg/mL among all analogs.•The most potent compounds with MIC < 40.96 μg/mL were screened toward HEK293T cells to test their cytotoxicity.•Docking studies of P35 at the active site of the chosen proteins Thymidylate Kinase (TMPK) from Mtb (PDB-1G3U), revealed the required interactions with the target site.
