Objective To investigate the effects of Pifithrin-μ on the proliferation of TSC2 mutant hepatocellular carcinoma cells. Methods The effect of Pifithrin-μ on cell proliferation was evaluated using CCK8 assay in multi-ple lines of Tsc2 mutant mouse embryonic fibroblasts(MEFs)and human hepatocellular carcinoma cells.Lentiviral infection was used to construct hepatocellular carcinoma cells with knockdown or overexpression of TSC2.The pro-tein expression of TSC2 and the downstream effectors of the mTOR pathway were detected by Western blot.Cellular sensitivity to Pifithrin-μ was detected by CCK8.The nude mouse subcutaneous transplantation tumor model was established for evaluation of drug efficacy in vivo.RNA sequencing was used for differential gene and pathway en-richment analysis, which was verified by Western blot and CCK8.Results Pifithrin-μ selectively inhibited the proliferation of Tsc2 mutated MEFs and hepatocellular carcinoma cells in a dose-dependent manner(P<0.05).Hepatocellular carcinoma cells with TSC2 knockdown showed increased sensitivity to Pifithrin-μ,whereas those with TSC2 overexpression were resistant(P<0.05).Pifithrin-μ significantly inhibited the growth of Tsc2 mutant transplantation tumors in nude mice(P<0.05).Pifithrin-μ downregulated the expression of Smad7 protein in Tsc2 mutant cells, resulting in elevated levels of p-Smad2/3 and over-activation of the TGF-β/Smad signaling pathway(P<0.05).The combination of each of the six cell death inhibitors with Pifithrin-μ revealed that reac-tive oxygen species inhibitor and TGF-β/Smad pathway inhibitor were able to reverse Pifithrin-μ-induced cell death in TSC2 mutant cells(P<0.05).Conclusions Pifithrin-μ selectively inhibits the proliferation of TSC2 mutant hepatocellular carcinoma cells. This effect is associated with the downregulation of Smad7 and the disrup-tion of the TGF-β/Smad signaling pathway balance.
Currently, the treatment of rectal cancer is primarily surgery-based comprehensive therapy. For patients with rectal cancer, accurate assessment of disease and effective prediction of prognosis are crucial steps in formulating individualized treatment plans and improving survival outcomes. Radiomics provides a novel approach for the further utilization of imaging data in rectal cancer patients, offering assistance in assessing disease and predicting patient prognosis. Based on relevant research reports, this article reviews the progress of radiomics in clinical diagnosis, treatment, and prognosis prediction for rectal cancer patients, focusing on the assessment of clinicopathological parameters (lymph node metastasis, tumor deposits, extramural vascular invasion, perineural invasion, distant metastasis) and the prediction of overall survival and disease-free survival.
Air pollution is a significant public health problem, and particulate matter, one of the important air pollutants, has attracted global attention for its impact on human health. Although a large number of studies have been conducted to investigate the impacts, previous studies of particulate matter usually focused on the diseases of respiratory and cardiovascular systems, and less attention was paid to the effects of exposure to particulate matter on renal system. This paper systematically analyzes the differences in the effects of exposures to particulate matter with different particle sizes on chronic kidney disease based on the data of a large number of cross-sectional studies and cohort studies, and introduces the influence of multiple effect modifiers on the relationship between exposures to particulate matter and chronic kidney disease. The findings provide a reference for reducing the burden of chronic kidney disease attribute to particulate matter exposure and evidence to support the precise prevention of chronic kidney disease.
Hemorrhoids are a common anorectal disease. However, public awareness regarding hemorrhoids is generally low, and misconceptions are prevalent. A considerable proportion of patients with hemorrhoids experience delayed diagnosis and treatment due to embarrassment or reliance on information from non-professional sources. This guideline was jointly initiated by the Medical Science Popularization Branch of the Chinese Medical Doctor Association, the Anorectal Surgeons Branch of the Chinese Medical Doctor Association, the Anorectal Science Popularization Group of the Medical Science Popularization Branch of the Chinese Medical Doctor Association, and the Anorectal Disease Professional Committee of the Chinese Medicine Education Association. Led by Professor Li Chunyu from The Fourth Affiliated Hospital of China Medical University, it was developed collaboratively by renowned national experts in the fields of anorectal diseases and science popularization. This article provides statements covering epidemiology, risk factors, clinical manifestations, prevention of hemorrhoids, management of hemorrhoids, and perioperative management of patients with hemorrhoids. It aims to serve as a reference for medical staff in conducting science popularization work related to the prevention and management of hemorrhoids, thereby contributing to the improvement of hemorrhoid prevention and management standards.
Pediatric anal fistula is a relatively common anorectal disease in children. Its pathogenesis is associated with multiple factors, including anatomical abnormalities, infection, and immunodeficiency.Early identification and standardized management are crucial for improving prognosis. This guideline was jointly initiated by the Medical Science Populariza-tion Branch of the Chinese Medical Doctor Association,Anorectal Surgeons Branch of the Chinese Medical Doctor Associa-tion,Anorectal Science Popularization Group of the Medical Science Popularization Branch of the Chinese Medical Doctor Association. It was led by Professor Li Chunyu from the Fourth Affiliated Hospital of China Medical University and devel-oped by a national panel of renowned experts in anorectal diseases and science popularization. This guideline systemati-cally addresses the epidemiology, anatomical composition and classification, clinical manifestations, diagnosis and differen-tial diagnosis, treatment, postoperative management, and prevention of pediatric anal fistula. It is intended to serve as a reference for clinical medical professionals in science popularization regarding prevention and treatment of this condition,to promote the standardization and normalization of popular science content,to enhance the uniformity of clinical prac-tice,to reduce complications and recurrence rates, and ultimately to improve the quality of life of affected children.
