Cadonilimab, recognized as the most rapidly developed Chinese drug in the field of cervical cancer, received marketing approval in China in 2022. We conducted a study to investigate the safety and efficacy of Cadonilimab, combined with and without chemotherapy or bevacizumab treatment, in patients with recurrent / metastatic gynecological malignancies. We retrospectively enrolled 60 patients with recurrent / metastatic gynecological malignant tumors, confirmed by pathology or cytology from July 2022 to December 2024. All patients received at least two cycles of Cadonilimab. The primary endpoints were the investigator-confirmed objective response rate and disease control rate according to RECIST v1.1. Secondary endpoints included progression-free survival and safety. Fifty-six patients were included in the efficacy analysis. In the study population, This study is a retrospective real-world study, although it lacks a control group. There were 16 first-line patients with advanced cervical cancer, ORR was 81.3
Background: Serum prostate-specific antigen (PSA) is a primary metric for diagnosis and prognosis of prostate cancer (PCa). Exposure to heavy metals, such as lead, cadmium, mercury, and zinc can impact PSA levels in PCa patients. However, it is unclear whether this effect also occurs in men without PCa, which may lead to the overdiagnosis of PCa. Method: Data on a total of 5,089 American men who had never been diagnosed with PCa were obtained from the National Health and Nutrition Examination Survey performed from 2003–2010. The relationship between serum PSA levels (dependent variable) and concentrations of lead (μmol/L), cadmium (nmol/L), and mercury (μmol/L) were investigated with dietary zinc intake being used as a potential modifier or covariate in a weighted linear regression model and a generalized additive model. A series of bootstrapping analyses were performed to evaluate sensitivity and specificity using these models. Results: Regression analyses suggested that, in general, lead, cadmium, or mercury did not show an association with PSA levels, which was consistent with the results of the bootstrapping analyses. However, in a subgroup of participants with a high level of dietary zinc intake (≥14.12 mg/day), a significant positive association between cadmium and serum PSA was identified (1.06, 95% CI, P=0.0268, P for interaction=0.0249). Conclusions: With high-level zinc intake, serum PSA levels may rise in PCa-free men as the exposure to cadmium increases, leading to a potential risk of an overdiagnosis of PCa and unnecessary treatment. Therefore, environmental variables should be factored in the current diagnostic model for PCa that is solely based on PSA measurements. Different criteria for PSA screening are necessary based on geographical variables. Further investigations are needed to uncover the biological and biochemical relationship between zinc, cadmium, and serum PSA levels to more precisely diagnose PCa.
Background MicroRNA (miRNA) plays an important role in nasopharyngeal carcinoma (NPC). We aimed to identify a microRNA that can help us understand the molecular mechanism and develop target therapy for NPC. Methods: MiRNA expression levels in 18 specimens of nasopharyngeal carcinoma were firstly analysed using quantitative real-time PCR (qRT-PCR). Then we further detected vascular endothelial growth factor A (VEGFA) protein with immunohistochemistry in 108 paraffin-embedded samples from the First Affiliated Hospital of Guangzhou Medical University (Guangzhou, China). The cell line Hone1, 5-8F, CNE1 and CNE2 were employed in transwell invasion assay, western blot, RT-PCR and dual luciferase assay. Results: We found miR-195-5p had a lower expression level in high metastasis potential NPC, compared with low metastasis potential NPC (p < 0.05). In in-vitro experiment, the over expressed miR-195-5p resulted in significant decrease in migration and invasion of NPC cells. In contrast, the down-regulation of miR-195-5p increased their migration and invasion. By using dual luciferase assay, we found a binding site between miR-195-5p and 3’-UTR of VEGFA. Through direct binding, miR-195-5p can suppress VEGFA in NPC cell. The results of immunohistochemistry showed that patients with high expression of VEGFA had a poor distance-metastasis free survival (p < 0.05). Conclusions: It was concluded that the expression level of miR-195-5p in patients with NPC was associated with metastasis by targeting VEGFA, which suggests that miR-195-5p is a potential biomarker of prognosis and molecular therapeutic target in NPC.
