Memorial (Russian: Мемориа́л, IPA: [mʲɪmərʲɪˈaɫ]) is an international historical and civil rights society that operates in a number of post-Soviet states. Founded in 1989, it focuses on recording and publicising the Soviet Union's totalitarian past, but also monitors human rights in Russia and other post-Soviet states.
Recurrent mutations in splicing factors (SFs) have been established as crucial drivers of tumorigenesis in several types of blood cancer and are also common in a variety of solid tumors. Mutations change the RNA-binding preferences of SFs, promote global splicing alterations, and often generate erroneous mRNAs that are then degraded by nonsense-mediated mRNA decay (NMD). Consequently, several critical genes linked to hematopoiesis are dysregulated, leading to blood cancer. Although the field has progressed considerably in identifying aberrant genes and affected pathways, effective therapies have not yet emerged. To address this key gap, we instigated a gene-specific targeted strategy by unlocking the regulatory network. As a proof of concept, we scrutinized a tumor suppressor gene, EZH2, which is a bona fide target in SRSF2 mutated cancer. We precisely defined splicing cis-elements in EZH2 transcripts and illustrated the dynamic choreography of regulatory proteins in the entire splicing and NMD catalytic pathways. We then designed antisense oligonucleotides (ASOs) targeting important regulatory sites. Our lead ASO successfully corrects aberrant splicing and NMD, restores the expression and function of EZH2, and partially rescues hematopoietic defects and cellular properties. Our study demonstrates that ASO pharmacology is an actionable strategy for clinical development, challenging the existing paradigms in SF mutated cancers.
In most of the cases Interventional Radiology techniques and therapies are proposed for the management of symptomatic soft tissue benign tumors responsible for pain and/or compression symptoms aiming to offer a curative intent by means of tumor necrosis with subsequent symptoms’ management and improvement of life quality. The ablative therapies include chemical, thermal and non-thermal approaches while, trans-arterial (chemo)embolization also has a distinct role. Adjunct ancillary techniques should be performed whenever necessary to increase efficacy and safety and avoid or reduce complications. The purpose of the current review is to identify the basis for treating soft tissue benign tumors with Interventional Radiology therapies, to offer a detailed review of them, to explain the expected outcomes and describe techniques for avoiding complications. Furthermore, a reflection upon future directions will be suggested.
Background: Mnemonic discrimination (MD) involves distinguishing new stimuli from memories of highly similar “lure” items or events, and is a putative indirect probe of dentate gyrus functioning. MD is impaired in the elderly and in individuals with hippocampal lesions, schizophrenia, major depressive disorder, and Alzheimer’s disease. The gold-standard MD test, called the mnemonic similarity task (MST), is rarely used in clinical research. We therefore aimed to validate a novel analysis method that extracts information about MD and recognition memory in widely clinically used recognition memory tests which do not have categorical distinctions between “lures” and “foils.”Methods: By fitting a logistic function to the relationship between stimulus interference and the probability of classifying a stimulus as novel, at the single participant level, we derived participant-level indices of MD (λ) and overall recognition memory performance (Δ). We applied the novel measures to MST data from two independent datasets (N=18; N=67). Using linear mixed-effects modelling, we sought to confirm that λ predicts the MST’s lure discrimination index (LDI), while Δ predicts the MST’s overall recognition memory index (REC). Results: Across both datasets, λ predicted LDI (β=0.76, 95% CI [0.62-0.91], p<0.001), but not REC (β=-0.06, 95% CI [-0.20-0.09], p=0.438), while Δ predicted REC (β=0.93, 95% CI [0.83-1.02], p<0.001), but not LDI (β=0.06, 95% CI [-0.03-0.15], p=0197). The λ and Δ indices were not correlated. Conclusion: Our novel measure accurately indexes MD, without correlating with overall recognition memory performance. Future studies should apply it to large clinical datasets with widely used recognition memory tests, such as the California Verbal Learning Test.
This study investigates the relationship between U.S. and Japanese interest rates, emphasizing their domestic and international effects and their influence on other key economic indicators. By analyzing 30 years of data, including interest rates, Consumer Price Index (CPI), Producer Price Index (PPI), and unemployment rates for both countries, the research uncovers a significant correlation between the economic shifts in the U.S. and Japan. The findings demonstrate the interconnectedness of these two major economies, where changes in one countrys interest rates often lead to measurable impacts in the others economic performance. This research underscores the critical role of coordinated policy decisions between major economies, such as the U.S. and Japan, in maintaining global financial stability. The results suggest that synchronized economic policies are essential for mitigating potential adverse effects on global markets, highlighting the need for international collaboration in monetary policy to sustain economic equilibrium in an increasingly interconnected world.
