
Three pentacyclic triterpenoids including one new guajavanoic acid (2) and two known obtusinin (1) and goreishic acid I (3) have been isolated from the leaves of Psidium guajava. The new constituent 2 has been characterized as 2alpha-hydroxy-3beta-p-E-coumaroyloxyurs-12, 18-dien-28-oic acid through 1H-NMR and 13C-NMR (broad band and DEPT). This is the first report of isolation of compound 1 and 3 from the genus Psidium.
A new triterpene named as alysinol 3alpha, 22beta-dihydroxyolean-12-ene [structure: see text] along with known compounds usnic acid, methyl 2,4-dihydroxy-3,6-dimethyl benzoate, 3-hydroxy benzoic acid, stigmasterol, poriferasterol and ursolic acid have been isolated from Alysicarpus monolifer. The structure of [structure: see text] has been established through spectroscopic techniques.
Six known ent-kaurene, a new ent-kaurane and a new pimarane diterpenes were isolated from Sideritis trojana. The structures of new compounds were determined as ent-7alpha-15beta,16beta-epoxykaurane (1), and ent-2alpha-hydroxy-8(14),15-pimaradiene (2) along with the known compounds siderol (3), sideridiol (4), 7-epicandicandiol (5), isocandol B (6), candol A acetate (7) ent-7alpha-acetoxykaur-15-ene (8) by IR, 1D and 2D NMR techniques and HRMS.
Danazol (17beta-hydroxy-17alpha-pregna-2,4-dien-20-yno-[2,3-d] isoxazole) (1) on fermentation with Fusarium lini, Aspergillus niger, and Cephalosporium aphidicola yielded 17beta-hydroxy-2-(hydroxymethyl)-17-alpha-pregn-4-en-20-yn-3-one (2) and 17beta-hydroxy-2-(hydroxymethyl)-17alpha-pregna-1,4-dien-20-yn-3-one (3), while the fermentation of 1 with Bacillus cerus yielded compound 2 only.
Aspergillus niger cultures monohydroxylate ilicic alcohol in C-3 in a cis position with respect to the methyl groups at C-4 and C-10, and trans position with respect to the hydroxyl group of C-4. Furthermore, Aspergillus niger cultures reduce ilicic aldehyde to its corresponding alcohol.
The petrol extract from mace of Myristica argentea Warb. afforded six phenylpropenes, three lignans, three neolignans and a dilignan, bis erythro-argenteane ( 4 ) or rel-(8R,8'S,8"S,8"'R)-5',5"'-bis(7-(3,4-methylenedioxyphenyl)-7'-(4'-hydroxy-3'-methoxyphenyl)-8.8'-lignane]. The last-named compound is a new natural product.
The microbial transformation of (+)-adrenosterone (1) by Cephalosporium aphidicola afforded three metabolites identified as androsta-1,4-diene-3,11,17-trione (2), 17beta-hydroxyandrost-4-ene-3,11-dione (3) and 17beta-hydroxyandrosta-1,4-diene-3,11-dione (4). The fermentation of 1 with Fusarium lini also produced metabolites 2 and 4, while the fermentation with Trichothecium roseum afforded metabolite 3. The structures of transformed products were determined by spectroscopic methods.
In the course of our studies on the chemical constituents of the leaves of Skimmia laureola, a new triterpene O-methyl cyclolaudenol (1) and a new coumarin, (+)-7-methoxy-6-(2'R-methoxy-3'-hydroxy-3'-methyl butyl) coumarin (2) were isolated. In addition five known coumarins, isogospherol (3), heraclenol (4), 5,8-dimethoxy coumarin-2H-1-benzopyran-2-one (5), 7-methoxy-6[2'-oxo-3'-methyl butyl] coumarin (6), and (+)-ulopterol (7) were also isolated for the first time from this plant. The structures were identified by spectroscopic studies and the stereochemistry at C-2' in compounds 3 and 4 were established by Horeau's procedure.
The effect of photo-stabilization of Azadirachtin-A (Aza-A) was examined when exposed to sunlight and ultraviolet light in the presence of four structurally different ultraviolet stabilizers namely 4-aminobenzoic acid, 2,4-dihydroxybenzophenone, 4,4'-dihydroxybenzophetione and phenyl salicylate. The percentages of Aza-A recovered at different time intervals from slides exposed to different light conditions with and without UV stabilizers as well as kinetic studies indicated that the addition of phenyl salicylate in methanolic solution of Aza-A (in 1: 1 mole ratio) provides the best photo-stabilization of Aza-A molecule among the four UV stabilizers studied.
Three saponins, craniosaponin A (1) and buddlejasaponins Ia (2) and I (3) were isolated from the n-butanol soluble fraction of Craniotome furcata for the first time. Among them, craniosaponin A (1) was identified to be a new compound. The structure of craniosaponin A was assigned mainly by spectral methods. A preliminary assay in vitro was applied to evaluate their cytotoxicity against several tumor cell-lines.
From the 95% ethanol extract of the Hong Kong liverwort, Marchantiapaleacea, guided by bioactivity directed isolation, two novel isoriccardinquinones A and B were obtained together with previously known marchantin C, isoriccardin C and phenanthrene derivative, 2-hydroxy-3,7-dimethoxyphenanthrene. The structures of the new compounds were established by high field spectroscopic methods, including 2D NMR techniques.
From the roots of Rumex crispus, two known anthraquinones and a new one together with a new anthrone were isolated and the structures of compounds 1-4 were elucidated by spectroscopic means. The singlet oxygen generation capacity was tested with 1,3-diphenylisobenzofuran (DPBF) for compounds 1-4.
A series of 3-substituted guaiazulene derivatives has been synthesized and their antioxidant properties were evaluated by monitoring their capacity for scavenging the stable free-radical DPPH. 3-Vinylguaiazulene was the most potent, possessing antioxidant activity superior than alpha-tocopherol. These derivatives were also moderate inhibitors of the proliferation of human promyelocytic leukemia cells.
The ethyl acetate soluble part of the chloroform extract of Pulicaria gnaphalodes belonging to the family Asteraceae afforded a new sesquiterpene-dimer of guaiane class named as gnapholide and anabsinthin of the same skeleton. The structures of both the compounds were elucidated with the aid of spectroscopic techniques including 2D NMR.
New triterpene, 3 -( trans -cinnamoyloxy)-19 -hydroxy-urs-12-ene ( 1 ) has been isolated from the methanolic fraction of Debregeasia salicifolia , along with uvaol ( 2 ), 3 ,19 -dihydroxy-urs-12-ene ( 3 ), ursolic acid ( 4 ), pomolic acid ( 5 ), pomolic acid methyl ester ( 6 ) and tormentic acid ( 7 ) reported for the first time from this species. The compounds ( 1 ), ( 3 ) and ( 6 ) showed significant antimicrobial activity. The structure elucidation was made with the help of extensive 2D NMR spectroscopic techniques.
3,4-Dihydroxy-7,8-dihydro-beta-ionone 3-O-beta-D-glucopyranoside 1 has been isolated from a methanolic extract of apple (Malus domestica) leaves by XAD-2 adsorption chromatography and subsequent purification by high speed countercurrent chromatography (HSCCC) and HPLC. Under acidic conditions this glycoside gives rise to a number of volatile compounds including 1.1,6-trimethyl-1,2-dihydronaphthalene (TDN) 2, which is known as an off-flavor compound in wine. In the course of the study, six additional glycoconjugates were isolated from apple leaves. i.e. roseoside 3, 3-oxo-alpha-ionol beta-D-glucopyranoside 4, benzoic acid beta-D-glucose ester 5, kaempferol 3-O-rhamnoside 6, (+)-syringaresinol beta-D-glucopyranoside 7 and 4-hydroxy-5-(3"-methyl-2"-butenyl)-benzoic acid methylester 3-beta-D-glucopyranoside 8. The latter compound has been isolated for the first time in nature. In order to avoid the 'kerosene-off-flavor' caused by TDN, the methanolic extract has been subjected to yeast fermentation. This treatment reduced the amount of TDN formed and resulted in an improved flavor impression of the methanolic extract.
In continuation of our interest in chemical modification of triterpenoids, the Willgerodt-Kindler reaction of a lupane type triterpenoid lupenone provided a novel dimerized product 2. Formation of 2 is associated with an unusual oxidative dimerization of lupenone under Willgerodt-Kindler reaction conditions. The structure 2 was confirmed by extensive analysis of spectroscopic data including ES-MS and 2D-NMR.
The 9-O-beta-D-glucopyranosides of (6R)-3-oxo-4-hydroxy-7,8-dihydro-alpha-ionol 1 and 3-oxo-5,6-epoxy-beta-ionol 2 were isolated from quince leaves. The glycosidic extract was obtained by XAD-2 adsorption and MeOH elution. After DCCC separation and flash chromatography, purification by high performance liquid chromatography was carried out. The novel quince leaf constituents were characterized as peracetates 1a and 2a.
From the aerial parts of Salvia aspera one new tri-nordammarane named amblyol ( 1 ) was isolated besides amblyone ( 2 ). An X-ray analysis was performed on compound 1 . In addition, three known neoclerodane diterpenoids were also isolated. The structures of these compounds were established by spectral and chemical methods. The presence of dammarane triterpenoids in a Mexican Salvia is described for the first time.
A novel metabolite related to the previously identified compound 3-O-methylfunicone (1) was isolated from the fungus Penicillium pinophilum and its structure elucidated by spectral analysis as 3-O-methyl-5, 6-epoxyfunicone (2).
