
Abstract The aim of the study was to determine the therapeutic efficacy o f simultaneous administration of GnRH and PGF2α in dairy cows with ovarian cysts. Ovarian cyst-affected dairy cows were divided into two experimental groups: 54 cows treated with GnRH and PGF2α, and 42 cows treated with GnRH alone, whereas 22 untreated cows served as the control group. Clinical response and reproductive performance were evaluated. The cumulative disappearance was better in treated cows than in the control group; however, there were no differences between the treatment groups (92.6; 95.2% vs. 72.3%). The mean interval from calving to conception was not significantly shorter (being so by 29 d) in the GnRH/PGF2α group than in the cows treated with GnRH alone (P > 0.05). The intervals from treatment to conception were also similar in these groups. The pregnancy rate in both treated groups was similar (62%) and higher than in the control cows (53%). In the cows with luteal cysts, the total pregnancy rate was higher in all experimental groups; however, only in GnRH-treated cows was this difference statistically significant (77.8% vs. 50.0%, P < 0.05). With time after parturition, the pregnancy rate decreased in all groups. In general, the cows treated with GnRH and PGF2α simultaneously displayed a good clinical response and slight improvement in reproductive performance compared to the single-therapy GnRH group; however, this was not fully convincing.
Abstract The effect of inflammatory bowel disease (IBD) on the density of galanin - immunoreactive (GAL-IR) nerve fibers was determined in the mucosa of canine duodenum, jejunum, and descending colon. Fiber density was evaluated by a single immunofluorescence method in biopsy specimens obtained from healthy dogs and patients with variable severity of the disease. The density of GAL-IR nerve fibers was determined by the semi-quantitative method by counting fibers in the field of view (0.l mm2). Fiber density was higher in dogs with moderate and severe IBD than in healthy animals. The results of the study suggest that GAL present in intestinal nerve fibers could play a role in the pathogenesis and development of canine IBD.
Abstract Fenpropathrin (FEN) was administered intraperitoneally in doses of 2.38 mg/kg, 5.9 5mg/kg, or 11.9 mg/kg b.w. to mice for 28 consecutive days. FEN did not significantly affect the activity of alanine transaminase (ALT) in the sera. Superoxide dismutase and glutathione peroxidase activities were significantly elevated in the liver after a 28-d exposure to moderate or highest doses of the pesticide. These results demonstrate that the 28-day exposure to FEN leads to an up-regulation of expression of antioxidant enzymes in response to an oxidative stress in mouse liver without causing a significant increase in ALT activity.
The immunoreactivity of haemagglutinin (HA) polypeptides of equine influenza virus was compared among the strains isolated in Poland, using H3 monoclonal antibody. A stronger signal in immunoblot reaction was observed for AJequi/Pulawy/2008 HA polypeptides compared to A/equi/Pulawy/2006, despite the fact that both strains are phylogenetically closely related and belong to Florida clade 2 of American lineage. The strongest signal, observed in the case of A/equi/Pulawy/2008, seemed to be connected with the presence of G135, I213, E379, and/or V530 instead of R135, M213, G379, and I530 present in A/equi/Pulawy/2006 HA sequence. This implies that point mutations within amino acid sequences of HA polypeptides of equine influenza virus may change their immunoreactivity even when they are not located within five basic antigenic sites.
Abstract The study was conducted on Polish Merino (143♀ and 39♂) and old-type Polish Merino sheep (176♀ and 61♂) in Brylewo flock (Wielkopolskie Province). The examined sheep were at the age of one year. Prion protein (PrP) genotype was determined in all animals. In both breeds four alleles (ARR, ARQ, AHQ, VRQ) were found. Highly significant effect of breed and insignificant impact of gender within breed was observed in regard to the frequency of occurrence of alleles and genotypes susceptible to classical scrapie in the sheep. Eight different PrP genotypes in Polish Merino and nine genotypes in old-type Polish Merino were identified. Very high frequency of ARR/ARR genotype in old-type Polish Merino and high frequency of ARR/ARQ genotype in Polish Merino were found, with relatively significant frequency of occurrence of the genotypes containing VRQ allele. In old-type Polish Merino, three animals (of both genders) had VRQ/VRQ genotype. Breeding work involving elimination of animals encoding valine at codon 136, and introduction of rams with ARR allele to the population increased the frequency of occurrence of ARR/ARR genotype and ARR allele in the population of old-type Polish Merino. To improve the distribution of the genotypes genetically resistant to scrapie in the flock of Polish Merino only rams with ARR/ARR genotype were left. This guarantees an increase in the frequency of occurrence of genotypes genetically resistant to scrapie in the offspring.
The aim of this study was to determine the correlation between lipophilicity and maximum residue limit (MRL) value specified for veterinary drugs in the fatty tissue of various animal species. The analysis was performed on a group of 73 compounds with different modes of action and MRL values determined for the fatty tissue of animals. Additionally, the logarithm of water/organic phase partition ratio (LogP) and the ratio of ionised and unionised substance in buffer with pH 7.4 (LogD(7.4)) were calculated. The main analysis was performed after the division of the whole group into six fractions. The linear correlation and regression analysis were used to determine the indirect relationship between the mean arithmetic value of LogP or LogD7.4 in selected fractions and related LogMRL of the drugs tested. The calculations revealed a linear correlation between fractioned lipophilicity and LogMRL values for the analysed compounds. The existence of indirect relationship between lipophilicity and MRL values determined for fatty tissue was confirmed.
The aim of the study was to identify the affinity of 10 Staphylococcus strains isolated from table chicken eggs to specific species. Preliminary analysis performed by API ID32 Staph test identified these strains as S. aureus, but they exhibited a negative reaction in the tube coagulase test. Thus, the analysed strains were initially characterised as Staphylococcus aureus-like (SAL). Further characterisation was performed by genotypic methods, using restriction fragment length polymorphism (RFLP) of the coagulase gene (coa) and sequencing of the gene rpoB. An attempt was also made to identify the isolated Staphylococcus strains by MALDI-TOF mass spectrometry. The results indicated that none of the strains tested belonged to the species S. aureus. The rpoB sequences of five isolates showed the highest sequence similarity to S. haemolyticus, three isolates to S. chromogenes, and one isolate to S. epidermidis. One strain (SAL4) remained unidentified in this analysis. The results obtained using mass spectrometry were comparable to those based on gene sequence analysis. Strain SAL4, which could not be identified by sequencing, was identified by MALDI-TOF as Staphylococcus chromogenes.
Abstract The therapeutic effects of Sidr honey on second-intention healing of contaminated full-thickness skin wounds in dogs were investigated. Povidone-iodine was used as a standard treatment and served as a control. Healthy adult (two-to-four-year-old) mongrels, comprising six dogs and two bitches, were divided into four equal groups in order to obtain multi-aged wounds at the end of the study. Four 2cm × 2cm full-thickness skin wounds were created on both sides of the back area of each dog under general anaesthesia adhering to aseptic technique. Contaminated wounds were then divided into two treatment groups: Group 1, Sidr honey treated (right side wounds) and group 2, povidone-iodine treated (left side wounds). All wounds were evaluated grossly daily at the time of treatment application and digitally photographed once every week. Images were analysed using ImageJ software. The parameters of unhealed wound area and length of advancing epithelium were obtained. The epithelialisation areas, percentage of wound area, and wound contraction rate were then calculated. No significant differences were found between the two treatment groups in any of the parameters studied. Overall, both honey treated and iodine treated wounds healed well within the time period of the study (28 d). However, the study showed a beneficial effect of Sidr honey on second-intention healing of full thickness contaminated wounds in healthy dogs and the effect was comparable to that of Povidone iodine.
The purpose of this study was to assess the possible influence of beavers on the contamination of lake water with zoonotic parasites Giardia duodenalis and Cryptosporidium spp., with respect to the risk to human health. A total of 79 water samples were taken around the habitats of beavers from 14 localities situated in the recreational Masurian Lake District (north-eastern Poland). Water was sampled in the spring and autumn seasons, at different distances from beavers' lodges (0-2, 10, 30, and 50 m). The samples were examined for the presence of (oo)cysts of zoonotic protozoa Giardia duodenalis and Cryptosporidium spp. by direct fluorescence assay (DFA) and by nested and real time PCR. By DFA, the presence of Giardia cysts was found in 36 samples (45.6%) and the presence of Cryptosporidium oocysts in 26 samples (32.9%). Numbers of Giardia cysts, Cryptosporidium oocysts, and summarised (oo)cysts of both parasites showed a significant variation depending on locality. The numbers of Giardia cysts significantly decreased with the distance from beavers' lodges while the numbers of Cryptosporidium oocysts did not show such dependence. The amount of Giardia cysts in samples collected in spring was approximately 3 times higher than in autumn. Conversely, a larger number of Cryptosporidium oocysts were detected in samples collected in autumn than in spring. By PCR, Giardia DNA was found in 38 samples (48.1%) whereas DNA of Cryptosporidium was found in only 7 samples (8.9%), Eleven Giardia isolates were subjected to phylogenetic analysis by restriction fragment length polymorphism PCR or sequencing which evidenced their belonging to zoonotic assemblages: A (3 isolates) and B (8 isolates). In conclusion, water in the vicinity of beavers' lodges in the tested region was markedly contaminated with (oo)cysts of Giardia duodenalis and Cryptosporidium spp., which confirms the potential role of beavers as a reservoir of these parasites and indicates a need for implementation of appropriate preventive measures to protect tourists' health.
Abstract The study was performed on six male chinchillas. The animals were anaesthetised with ether and the anaesthesia was deepened with nembuthal injected intraperitoneally. The chinchillas were then transcardially perfused with 0.4 L of 4% buffered paraformaldehyde, and testes, epididymides, and vasa deferentia were collected. The tunica albuginea from one testis from each chinchilla was stained as whole-mount preparation. The tissues were cut into 12 μm-thick cryostat sections, and processed for double-immunofluorescence method. In all organs studied, the most abundant nerve fibres were dopamine β hydroxylase positive (DβH+). Some of them contained neuropeptide Y (NPY). Sporadically NPY-positive-only nerve fibres were found. Single DβH+ nerve terminals contained also galanine. Small numbers of the nerve fibres supplying studied organs were stained for substance P (SP) and calitonin gene related peptide (CGRP). Almost all SP+ fibres were also CGRP+, whereas single CGRP+ nerves were SPimmunonegative. Some nerve terminals were immunoreactive to vesicular acetylcholine transporter and vasoactive intestinal peptide. The organs studied were innervated unevenly. The highest density of the nerves was found in the areas of the tunica albuginea adjacent to the mesorchial border of the testis and their number gradually decreased towards the free border of the gonad. None of the vascular tissue of the testicular parenchyma was free of the nerve fibres, except sporadically encountered DβH+ nerves which supply seminiferous tubules. Within the head of the epididymis a moderate number of nerve terminals were found, but in the body and tail of the organ the number of nerves gradually increased. The vas deferens was supplied with very numerous nerve fibres. There were no differences in the density of the innervation between the funicular and abdominal part of the vas deferens.
The paper describes avian tuberculosis in a captive bred cassowary. A two-and-a-half-year-old bird was obtained by a Polish zoo in 2010 from the Netherlands under conditions compliant with the recommendations of the European Association of Zoos and Aquaria. Despite being of small size for the age, the bird appeared healthy and showed no signs of the disease until the day when it was found recumbent in its pen. Later on it was euthanised due to lack of treatment possibilities. Pathological changes typical of avian tuberculosis were found in the liver and spleen. Mycobacterium avium ssp. avium was cultured from both organs.
The aim of the study was to estimate the current prevalence of E. multilocularis in selected populations of red foxes in Poland and to evaluate the changes in prevalence of this parasite by comparison with the results obtained in the same area during earlier surveillance. The investigations were performed in the area of four Polish provinces: 2 eastern/south-eastern (Lubelskie and Podkarpackie) and 2 south-western (Slaskie and Opolskie). Five hundred red foxes coming from the investigated areas were examined between 2013 and 2014 to estimate the current situation in selected provinces. Moreover, 550 red foxes from the same areas examined between 2007 and 2013 were used for comparison of differences in E. multilocularis prevalences in time. Intestines were examined with the use of the sedimentation and counting technique. Among 500 foxes examined in the current study, 118 were positive for E. multilocularis. There were differences in prevalence between individual provinces: Podkarpackie Province - 54.6%, Lubelskie Province - 18.9%, Slaskie Province - 11.7%, and Opolskie Province - 3.9%. Statistical analysis demonstrated that in most cases there were no differences in prevalence between the current results and the results from previous studies. Only in Opolskie Province was a statistically significant increase observed between 2010 and 2014. A stable degree of infection in the region with high prevalence of this parasite was demonstrated. However, a significant increase in the region with very low prevalence of E. multilocularis points out the necessity to monitor this infection during the coming years to control the progress of the disease.
The paper describes a microbiological method for the detection of antibacterial substances in feedingstuffs. The method allowed detection of the main antibiotic groups, including tetracyclines. In 2013-2014, a total of 171 feed samples were analysed to determine antibacterial substances. Among the analysed samples 84 (49.1%) were suspected to contain tetracyclines. Out of the 84 feeds analysed using chromatography, 28 (33.3%) contained undeclared tetracyclines, which were identified at concentrations ranging from 0.32 mg kg(-1) to 48.98 mg kg(-1).
Abstract The study was performed on nasal swabs, tracheal samples, and sera obtained from young beef heifers aged between 6 and 12 months, from farms in eastern and south-eastern Poland. The samples were evaluated using bovine herpesvirus 1 (BHV-1) ELISA kits (ELISA BHV1 antibody and ELISA BHV1 antigen) and PCR. Among all the animals examined, 37 (32.2%) were positive in the ELISA BHV1 antigen test. The presence of BHV-1 was confirmed by PCR in 42 (36.5%) animals. In the ELISA BHV1 antibody test, 39 (33.9%) seropositive animals were identified. The presence of BHV-1 positive samples was observed in all the examined breeds of young cattle. There were no significant differences (P ≤ 0.05) in BHV-1 positive samples. The results indicate that the incidence of BHV-1 infections in feedlot cattle herds studied was 32.2%-36.5%, which suggests that preventive measures should be implemented in order to limit transmission of the virus.
The paper presents the results of testing eggs for the content of dioxins and polychlorinated biphenyls (PCBs), determination of the sources that caused the presence of high concentrations of these compounds which exceeded the acceptable contamination levels, and assessment of consumer health risk caused by the consumption of eggs with excessive contents of investigated compounds. In about 9% of free range eggs and 17% of organic eggs, the content of PCDD/Fs was two- or three-fold higher than the acceptable limits, and in some samples the concentration of investigated compounds exceeded the maximum concentration levels. Based on the profile of the compounds, it was confirmed in several cases that their main source was the soil or unsecured refuse. The consumers of eggs and meat produced under these conditions constitute the risk groups, and their dioxin and PCB intake may exceed toxicological reference values.
Abstract A liquid chromatography – tandem mass spectrometry (LC-MS/MS) method for the determination of oxytetracycline (OTC), 4-epi oxytetracycline (4-epi OTC), tetracycline (TC), 4-epi tetracycline (4-epi TC), chlortetracycline (CTC), 4-epi chlortetracycline (4-epi CTC), doxycycline (DC), minocycline (MINO), methacycline (META) and rolitetracycline (ROLI) residues in muscles was developed. The procedure consisted of an oxalic acid extraction followed by protein removal with trichloroacetic acid. Further solid phase clean-up on polymeric (Strata X) reversed phase columns was performed to obtain an extract suitable for LC-MS/MS analysis. The tetracyclines were separated on a C 18 analytical column with mobile phase consisting of 0.01% formic acid in acetonitrile and 0.01% formic acid in water in gradient mode. The method was validated according to the Commission Decision 2002/657/EC. The recoveries of all target compounds were 91.8% – 103.6%. The decision limits were from 109.0 to 119.8 μg/kg and detection capability varied within the range of 122.2 to 137.6 μg/kg, depending on the analyte.
Abstract The aim of the study was to identify magnetic resonance imaging (MRI) sequences that contribute to a quick and reliable diagnosis of brachial plexus tumours in dogs. The tumours were successfully diagnosed in 6 dogs by the MRI with the use of SE, FSE, STIR, Turbo 3 D, 3D HYCE, and GE sequences and the gadolinium contrast agent
The aim of this study was to assess the effect of thymus factor X (TFX-Jelfa) treatment in hypoestrogenic female rats for the purpose of decreasing skin impairments. Ovariectomised rats were used as a model. The histopathological analysis of the skin after TFX-Jelfa treatment demonstrated that the epidermis was thicker and more desquamated, without deep wrinkles or hypersecretion in comparison to the skin of animals only castrated and not treated with TFX-Jelfa. Collagen and elastic fibres were arranged more uniformly in the dermis and there were numerous fibroblasts, hair follicles, and small vessels. Ultrastructural analysis showed keratinocytes without degenerative changes and the proliferation of lymphatic and dendritic cells in the skin. The results indicate that thymus extracts can have beneficial effects on skin aging, which is often accompanied by hormonal perturbances.
The aim of study was the preliminary evaluation of the occurrence of methicillin-resistant Staphylococcus aureus (MRSA) and extended spectrum beta-lactamases (ESBL) producing Escherichia coli in 650 milk and inflammatory secretions from cows with clinical or subclinical mastitis. One millilitre of the sample was added to Mueller-Hinton broth supplemented with 6.5% NaCl, Tryptone Soya Broth with cefoxitin and aztreonam, and then to MRSA ID agar. Presumptive MRSA colonies were analysed for the presence of mecA gene. Parallel to MRSA identification, the samples were incubated in buffered peptone water, lauryl tryptose broth and McConkey agar supplemented with cefotaxim for ESBL-producing E. coli isolation. These bacteria were identified using APl Rapid 32 E and the ability of ESBL production was initially established using disc test D68C and confirmed by MIC technique using Sensititre ESBL plates. The primers (blaCTX, blaTEM, blaSHV, and blaCMY-2-group) for the detection of some of the genes encoding ESBL production were used. The 45 strains of S. aureus with mecA gene and 41 strains of E. coli with blarTEM gene were detected.
Abstract A 3-year-old female fallow deer was subjected to the necropsy and virological testing, due to a suspected infectious disease in the herd of farmed deer in the Southeastern region of Poland. The animal was found negative for the presence of BVDV, BoHV-1, BTV, and EHDV antibodies and BVDV antigen. The toxicological examination did not reveal any coccidiostats, mycotoxins, rodenticides, carbamate pesticides, and organophosphorus pesticides. The flukes found during postmortem examination were first characterised microscopically as Fascioloides magna and later their identity was confirmed by PCR and sequencing. The autopsy revealed lesions characteristic for F. magna infection, including different size cystic spaces in the liver, filled with brownish mucous fluid and flukes, and black pigment covering the surface of parietal and visceral peritoneum with the highest concentrations localised next to the liver. The changes observed in the liver tissue were typical of liver cirrhosis. The results demonstrated that in Poland, where the cervid farming is developing dynamically, the problem of fascioloidosis is present and may probably exert a significantly negative influence on the productivity of such farms if no antiparasitic treatment is performed.
