This study investigated the prevalence, clinicopathological features, and prognostic significance of 11q aberrations in MYC-rearranged large B-cell lymphoma (LBCL) without BCL2 and BCL6 rearrangements. We retrospectively analysed 22 patients with LBCL (20 with DLBCL and 2 with HGBCL) diagnosed between 2015 and 2025. MYC rearrangement and the absence of both BCL2 and BCL6 rearrangements were confirmed by FISH. 11q status was assessed using dual-colour 11q23.3/11q24.3 probes, and correlations with clinicopathological parameters were analysed. 11q aberrations were detected in 9 of 20 patients with DLBCL (45
Lysine-specific demethylase 1 (LSD1/KDM1A) regulates hematopoietic differentiation and leukemic stem cell programs, making its inhibition a potential therapeutic strategy for hematologic malignancies. We systematically reviewed the clinical efficacy, safety, combination approaches, and biomarkers of LSD1 inhibitors in hematologic malignancies. PubMed/MEDLINE, Scopus, and Web of Science were searched from inception through 31 May 2026 for clinical trials evaluating pharmacologic LSD1 inhibition in hematologic malignancies. Eligible full-text reports and conference abstracts were screened, and study, treatment, efficacy, survival, safety, and biomarker data were extracted. Owing to substantial clinical and methodological heterogeneity, findings were synthesized narratively. Thirteen early-phase trials involving 456 enrolled participants evaluated five inhibitors: iadademstat, bomedemstat, tranylcypromine, GSK2879552, and seclidemstat. In R/R AML, monotherapy and ATRA-based combinations generally produced modest responses, whereas newly diagnosed or molecularly selected cohorts receiving iadademstat-based combinations reported higher response rates. However, differences in patient populations and active treatment backbones preclude attributing these cross-study differences to LSD1 inhibition itself. In myeloproliferative neoplasms, bomedemstat demonstrated blood count control and variable improvements in spleen volume, symptoms, marrow fibrosis, and driver mutation burden. Among studies with explicit denominators, study-level rates of any-grade treatment-related adverse events ranged from 72
Despite the rapidly evolving landscape, allogeneic transplant still plays a role in acute lymphoblastic leukemia. Relapse post-transplant is not uncommon, leading to dismal prognosis. Donor lymphocyte infusions efficacy in this setting remains controversial. In this retrospective study, we report the use of DLIs in a cohort of 43 ALL (30 B- 13 T-) patients, treated at our Center between 2012 and 2024. 2y-OS, EFS and GRFS were 69.7
Gastric mucosa-associated lymphoid tissue (MALT) lymphoma is an antigen-driven malignancy in which Helicobacter pylori (H. pylori) eradication achieves remission in most localized cases, yet the genomic underpinnings of biological heterogeneity across H. pylori subgroups remain poorly defined. We performed targeted next-generation sequencing on formalin-fixed paraffin-embedded tumor tissue from 28 patients with stage IE gastric MALT lymphoma, classified as H. pylori-negative (HPN; n = 8) or H. pylori-positive (HPP; n = 20). Recurrently altered genes were grouped into three functional categories: epigenetic and transcriptional regulatory genes, NF-κB signaling-related genes, and immune-related genes. Overall mutational burden was low (median: 1 altered gene per case; range, 0–9), with PTEN, KMT2D, and TET2 as the most frequently mutated genes, distributed across both subgroups. Across the predefined functional categories, epigenetic/transcriptional regulatory alterations were more frequent in HPN cases (62.5
Allogeneic hematopoietic stem cell transplantation (allo-HSCT) plays an important role in achieving long survival in adult T-cell leukemia/lymphoma (ATLL). Donor-derived hematopoietic cells can become infected with human T-cell leukemia virus type I (HTLV-1) following allo-HSCT from HTLV-1-negative donors; however, subsequent development of de novo ATLL of donor cell origin in this setting remains exceedingly rare. We encountered a 50-year-old woman who developed de novo unfavorable chronic-type ATLL of donor cell origin merely 2 years after receiving allo-HSCT from an HLA-matched, HTLV-1-negative sibling donor. Although she initially achieved complete remission following allo-HSCT, her clinical course was complicated by refractory acute graft-versus-host disease (GVHD). Consequently, she received additional prolonged immunosuppressive therapies, including ruxolitinib and mesenchymal stem cells, but adequate disease control could not be achieved. Approximately 2 years after allo-HSCT, a small proportion of atypical lymphocytes was recorded in her peripheral blood. HTLV-1-infected cell analysis using flow cytometry and Southern blotting analysis of the atypical lymphocytes revealed the development of ATLL. The patient’s disease status was considered to be at high risk of acute transformation; however, peripheral blood chimerism revealed complete donor type. A proviral load of 80.6 copies/100 cells was recorded alongside an expansion of multiple small clones and a dominant clone that accounted for 26.1
