Choanoflagellates are the only non-animals to contain key homologs of voltage-gated channels required for generating spontaneous action potentials. Action potentials activate high voltage-gated CaV channels which mediate neurotransmitter secretion at presynaptic nerve terminals (CaV2.1, CaV2.2), neuro-endocrine cells (CaV2.3) or sensory ribbon synapses associated with hearing (CaV1.3) and vision (CaV1.4). Cav1 channels in T-tubules align with ryanodine receptors of opposing sarcoplasmic reticulum (SR) membranes to mediate skeletal (CaV1.1) and cardiac (CaV1.2) muscle contractions.
