Objective:To construct hepatocellular carcinoma (HCC) cells lines that can mimic sub-lethal heat shock (HS) in vitro and orthotopic HCC mouse model that received incomplete thermal ablation.Methods:The suspension H22 cells were grown in 6-well plates, and the medium was replaced with fresh complete medium 8 hours before heat treatment. Then, the cells were divided into 4 groups for constant temperature water bath at 37°C, 42°C, 47°C, 50°C for 15 min, respectively. After the heat treatment was completed, the cells were maintained in a 37℃ incubator until detection. The phenotype of epithelial-mesenchymal transition (EMT) was detected by Western blot 48 hours later, and the cell viability was detected by Cell Counting Kit-8 (CCK-8) kit. The liver of mice was exposed by laparotomy along the abdomen midline and H22 cells (5×106) were injected through the hepatic capsule to conduct orthotopic HCC model. When the longest diameter of the tumor reached approximately 8 mm, the mice were randomly divided into complete ablation group (cMWA), incomplete ablation group (iMWA) and Control group treated with sham operation. Microwave ablation along the long axis of tumor was performed to establish the incomplete thermal ablation model of orthotopic HCC. MRI was performed for tumor measurement and liver specimens were collected for histopathological assessment at day 14 after microwave ablation.Results:14 daysafter inoculation of H22 cells, the tumors reached average tumor diameter of 9.32 ± 0.83 mm which was predetermined to be suitable for microwave ablation. 14 days after microwave ablation, the average tumor diameter reached 12.93 ± 1.51mm in iMWA group, which were significantly bigger than preoperative period. In addition, immunofluorescence staining showed a large number of infiltrated neutrophils in the residual tumor of iMWA group. Western blot was used to observe the change in EMT-phenotype, results showed that the expressions of Vimentin and α-SMA were up-regulated and the expressions of E-cadherin were down-regulated.Conclusions:HCC cells lines that can mimic Sub-lethal heat shock (HS) in vitro and orthotopic HCC mouse model that received incomplete thermal ablation were successfully constructed. There was substantial staining for neutrophils infiltration after incomplete ablation. Incomplete ablation may allow HCC cells to become more prone to migrate and to become more invasive.
Observational studies on the prognostic role of preoperative serum carcinoembryonic antigen (CEA) in non-small cell lung cancer (NSCLC) are controversial. Electronic databases updated until June 1, 2014 were searched to find relevant studies. A meta-analysis was conducted with eligible studies which quantitatively evaluated the relationship between preoperative serum CEA level and survival of patients with NSCLC. Survival data were aggregated and quantitatively analyzed. We performed a meta-analysis of 16 studies (n = 4,296 patients) that evaluated the correlation between preoperative serum CEA level and survival in patients with NSCLC. Combined hazard ratios suggested that preoperative serum CEA overexpression was associated with poor prognosis of overall survival (OS) (hazard ratio (HR) = 2.28, 95 % confidence interval (CI) 2.24–2.31) in patients with NSCLC. Meanwhile, for p-stage I NSCLC, the HR (95 % CI) was 1.98 (1.73–2.15). In the stratified analysis by patient source, significant risks were found among Asians and non-Asians. However, significant heterogeneity was observed among all studies. In the present meta-analysis, preoperative serum CEA overexpression indicates a poor prognosis for patients with NSCLC.
Objective To perform a meta-analysis to evaluate the association between the interleukin-8-251T/ A(rs4073) gene polymorphism and lung cancer risk. Methods A literature search at PubMed was performed to identify case-control study articles published before December 1st, 2013. The Stata11.0 software was used to calculate the pooled odds ratios(OR) with 95% confidence intervals(CI). And the analysis was stratified by Asian and non-Asian populations. Results Totally six articles met our inclusion criteria. The included studies yielded a total of 3 265 lung cancer cases and 3607 controls. For the homozygous A/ A and A allele carriers(T/ A + A/ A), the pooled ORs for all studies combining 3 265 cases and 3 607 controls were 1. 03(95% CI =0. 92-1. 14, P=0. 235 for heterogeneity) and 1. 07(95% CI =0. 96-1. 19, P=0.245 for heterogeneity) when compared with the homozygous wild-type genotype(T/ T). When the analysis was stratified by Asian and non-Asian populations, significant risks were found among Asians for both the A allele carriers(OR = 1. 48, 95% CI = 1. 04-2. 11, P = 0. 030 for heterogeneity) and the homozygous A/ A(OR=1. 35, 95% CI =1. 02-1. 92, P=0. 090 for heterogeneity) individuals. However, no significant associations were found in non-Asian populations using any of the genetic models. Conclusion This Meta-analysis suggests that the interleukin-8-251 allele confer an increased risk for the development of lung cancer among Asians.
