Botanically, there are two main types of tea i.e. China and Assam type, predominant in China and India, respectively. Different chromosome-level assemblies of China type tea genomes have been reported recently but none for Assam type. Thus, in the present study, genome assembly of most popular Assam type Indian tea cultivar, TV 1 was decoded at chromosome-scale and analyzed to understand the domestication, variations, and evolutionary differences between China and Assam type teas. Whole genome duplication (WGD) study has confirmed close collinearity and recent WGD events between Assam type teas from India (Camellia assamica) and China (C. sinensis var. assamica). Single copy gene-based study has confirmed divergence of TV 1 from China type tea (C. sinensis var. sinensis) approximately 5.5 MYA, and SNP-based analysis of 150 genotypes across the world supports their independent domestication. Moreover, 512 genes in TV 1 were found positively selected during domestication events between caffeine and non-caffeine groups. At 93 fusion genes per species per million years rate, 400 fusion genes were found with no shared recent fusion events between China and Assam type tea. Secondary metabolite profiling has yielded several metabolic compounds such as caffeine, theophylline, and catechins. Introduction:This in vitro study aimed to compare the load-to-failure values of two different ceramic veneers (IPS e.max Press [Ivoclar Vivadent] and Vintage lithium disilicate [LD] press [Shofu]) with incisal preparation designs under standardized conditions. Materials and Methods:Twenty-two intact extracted maxillary incisors were selected and divided randomly into two groups (n = 11). The veneer tooth preparation was standardized in both groups where a butt joint incisal preparation was done along with chamfer margin. Group 1 included veneers made from IPS e.max Press (Ivoclar Vivadent) and Group 2 included veneers fabricated from Vintage LD Press (Shofu). Veneers were luted to their respective abutment teeth using standardized bonding protocols and resin cement for both groups. Later, every specimen was loaded to failure utilizing a universal testing machine, and the outcomes were noted in Newtons (N). Results:The mean load-to-failure value obtained for Group 1 (IPS e.max Press) was 1386.46 N while that obtained for Group 2 was 1777.07 N. Statistically significant difference was found in this intergroup comparison (P = 0.006). Conclusion:The load-to-failure value of Vintage LD Press veneers (Shofu) was greater than that of IPS e.max Press veneers (Ivoclar Vivadent).
OBJECTIVE:Gallbladder carcinoma is the most frequent biliary tract carcinoma with over all very poor prognosis. Epidermal growth factor receptor (EGFR) is known to be involved in carcinogenesis and overexpressed in various malignancies including head and neck, breast, lung and colon carcinomas. This study was done to explore the expression of EGFR in gallbladder carcinoma cases in the north Indian population so that it may be used as a therapeutic target in these patients.MATERIALS AND METHODS:59 cases of gallbladder carcinoma diagnosed by histopathological examination were included in study. Expression of EGFR was seen by immunohistochemistry method on histopathology slides.RESULTS:Out of 59 gallbladder carcinoma cases 46 (78%) were female and 13 (22%) were male with female to male ratio of 3.54:1. Mean age was 51.71±11.32 years. On histopathological examination 51 (86.4%) cases were conventional adenocarcinoma, 2 (3.4%) adenosquamous carcinoma, 2 (3.4%) mucinous adenocarcinoma, 2 (3.4%) papillary adenocarcinoma, 1 (1.7%) signet ring cell carcinoma and 1 (1.7%) squamous cell carcinoma histological subtypes. EGFR expression was present in 31 (52.5%) of gallbladder carcinoma cases and strong EGFR expression was significantly associated with poor differentiation of tumour.CONCLUSION:In our study EGFR was positive in the majority of gallbladder carcinoma cases. There was inverse correlation between differentiation of tumor and EGFR expression. Strong EGFR expression was significantly higher in poorly differentiated tumors compared to well differentiated tumors suggesting its role in prognosis. This also suggest that EGFR might have a role in tumor progression and aggressiveness. Therefore, EGFR have potential to be used as therapeutic target in significant number of patients. More larger sample studies are required to confirm our findings. EGFR may be further studied as therapeutic target in clinical trials in the Indian population to improve morbidity and mortality of gallbladder carcinoma patients.KEY WORDS:EGFR Expression, Gallbladder Carcinoma, Immunohistochemistry, Targeted Therapy.
Background: Gallbladder carcinoma (GBC) although rare is most frequent malignant neoplasm of biliary tract system and sixth most common malignancy of digestive tract. GBC is more common in females and there are studies which show expression of estrogen receptor (ER), progesterone receptor (PR), and human epidermal growth factor receptor-2 neu (HER2/neu) in GBC suggesting possible molecules for targeted therapy, but results are inconsistent. Aims and Objectives: The aim of this study was to find out expression of ER, PR, and HER2/neu in GBC in North Indian population and their possible association with clinicopathological features. Materials and Methods: A total 59 resected cases of GBC diagnosed by histopathological examination were included in the study. Expression of ER, PR, and HER2/neu was accessed by immunohistochemistry method and correlated with various clinicopathological features. Results: ER expression was absent in all GBC cases. PR expression was present in only one case. Positive expression of HER2/neu was present in 13 (22%) cases, in which 12 cases were of conventional adenocarcinoma and one case was of papillary adenocarcinoma. Well and moderately differentiated tumor had significantly higher HER2/neu expression as compared to poorly differentiated tumors (P = 0.001). Pre-obese patients had significantly higher HER2/neu expression as compared to non-obese patients (P = 0.008). Conclusion: In our study, there was no expression of estrogen and PR in GBC in North Indian population. Although small in number, there is a subset of patients who overexpress HER2/neu receptor that may benefit from targeted therapy.
Aim:This study aimed to evaluate and compare the impact of different bleaching agents on the fracture resistance of endodontically treated teeth when using either GC Fuji type 2 glass ionomer cement (GIC) or Shofu Glass Ionomer RX EASE as intraorifice barriers (IOB). Materials and Methods:A total of 80 single-rooted human maxillary central incisors were prepared and obturated. Three millimeters of gutta-percha was then removed from the orifice. The specimens were divided into two primary groups based on the type of IOB material used: GC Fuji type 2 GIC and Shofu Glass Ionomer RX EASE (n = 40). Each group was further divided into four subgroups based on the bleaching agent used: Carbamide peroxide (CP) 37%, sodium perborate (SP), hydrogen peroxide (HP) 35%, and distilled water used as the control (n = 10). The teeth were subjected to fracture resistance testing. Results:The study found that the order of root fracture resistance was control > CP > SP > HP. There was no statistically significant difference in fracture resistance between GC Fuji type 2 GIC and Shofu Glass Ionomer RX EASE when used as IOB materials. Conclusion:The study concluded that the choice of bleaching agent significantly affects the fracture resistance of endodontically treated teeth. It was observed that fracture resistance is lowest with HP, followed by SP and CP. Both GC Fuji type 2 Glass Ionomer and Shofu Glass Ionomer RX EASE are effective as IOB.
Introduction: Cluster of Differentiation 9 (CD9), known as Motility Related Protein (MRP-1) regulates cell adhesion, motility, migration and proliferation. Many studies have stated conflicting results on prognostic significance of invasive breast carcinomas with CD9 expression that had performed on tumour tissues. Aim: To assess the inter-relationship of CD9 expression of tumour cells and stromal immune cells in breast carcinoma with clinicopathological parameters which include age, tumour size, grade, histological type, lymph nodes, tumour staging and molecular classification. Materials and Methods: An observational prospective (2020 July to 2021 June) and retrospective (2019 October to 2020 June) study was done in 71 cases of resected primary invasive breast carcinoma over a period of one and half year at Sri Devaraj Urs Medical College, Karnataka, India. Immunohistochemistry (IHC) staining was done using CD9 antibody. Tumour cells (T-CD9) expression was evaluated by Immunoreactivity scoring (IS= Intensity score × Extent of staining). The stromal cells (S-CD9) expression was evaluated by Percentage (%) of stromal area occupied by CD9 stained immune cells. Chi-square test was used as test of significance for qualitative data. The p-value of <0.05 was considered as statistically significant. Results: Out of 71 cases, T-CD9 expression was noticed in 40 cases (56.34%) and IS <4 considered as negative was observed in 31 cases (43.66%). However there was no association with age, tumour size, grade and molecular markers for the expression of both T-CD9 and S-CD9. Human Epidermal growth factor receptor 2 neu (HER2neu) negative was associated with T-CD9 expression (p-value=0.05). Hence CD9 can be used as prognostic marker for Her2neu negative cases. Conclusion: CD9 expression was not significantly associated with tumour cells (T-CD9) and stromal cells (S-CD9) in breast carcinoma cases. However it was significantly associated with Her2neu negative tumour cells. T-CD9 showed more positivity in Luminal A followed by Triple negative whereas S-CD9 showed more positivity in Luminal B. CD9 did not show association with any parameters except Her2neu negative.
Coconut testa and tender coconut water (TCW) are by-products of coconut processing industries which are rich in natural phytonutrients, if not exploited properly it may add up to environmental pollution. In the current study, phenolic concentrate (PHE) from coconut testa and tender coconut water (TCW) concentrates were examined for their effect on high fat fed C57BL/6 mice at the level of 50 & 100 mg and 500 & 1000 mg/kg body weight respectively. Results showed increasing body weight in high fat fed animals when compared to starch fed control (ND/SFD) group. Treatment with concentrates of PHE and TCW reduced their body weight dose dependently. Lipid profile like triglycerides, cholesterol and LDL levels were significantly decreased, whereas HDL levels increased, indicating its health beneficial effect. Catalase, SOD, GPx, TBARS in tissues, analysis of OGTT, serum insulin levels, advance glycation and atherogenic protection were augmented at different levels in the treated groups. In conclusion, phenolic concentrate prepared from the coconut testa and tender coconut water concentrate revealed their hypolipidemic property which can be exploited further in the development of health foods.
Guanine-rich DNA sequences may fold back into non-canonical four-stranded secondary structures termed as Gquadruplexes. The role of G-quadruplexes has already been well established in different diseases like cancer, neurological and viral disorders etc. Also, several small molecules have been reported, which can influence the involvement of G-quadruplexes either through stabilization or destabilization in the cellular environment. Growing statistics have associated G-quadruplex assemblies to a discrete biological process in vivo, including DNA replication, transcription, genomic stability, and epigenetic regulation. DNA G-quadruplex existence in human telomere is well recognized attractive target for anticancer drugs. G-quadruplex-interactive ligands have been known to prevent telomerase access as well as telomere capping. To the best of our understanding, the role of G-quadruplexes in virology, neuropharmacology, cancer progression and its treatment has not been discussed on a single platform till date. This review aims to enhance our knowledge regarding these magical sticky quadruplex structures, which have been quite significantly proved to be the part of many cellular processes along with their established in vivo existence. Understanding regarding stabilizing or destabilizing ligands of these multistranded guanine quadruplex structures might be proved as the facilitator of drug discovery process for many incurable diseases in future.
Electrochemical biosensors (an integrated system of biological constituent and physiochemical detector) have attracted plenty of attention due to its high precision, and importance in clinical diagnosis. In electrochemical biosensors, redox indicators like methylene (MB) blue are used to detect the electrical changes, which by intercalating between DNA base pairs, can produce an intense redox signal on the surface of ssDNA modified electrode. This study reports the interaction between calf thymus DNA (ctDNA) and new methylene blue (NMB), having almost identical basic skeleton to MB. Various techniques like UV-Visible, thermal melting, fluorescence, circular dichroism (CD) spectroscopy and molecular docking have been utilized. Hypochromism and a red shift in UV-Vis spectra revealed the intercalation binding mode for DNA-NMB complex. Nearly identical binding constants were calculated using UV-Visible and fluorescence spectroscopy. The calculated thermodynamic parameters like change in enthalpy (ΔH°) and entropy (ΔS°) were found to be -6.11×104 and -128.96 JK-1mol-1 at 290 K respectively. CD revealed a change into more compact B-DNA conformation after binding with NMB. These studies suggest that intercalation mode, hydrogen bonding, and van der Waals forces might be responsible for DNA-NMB interaction. This work might further facilitate our understanding about DNA-drug interactions for utilizing them for medicinal purposes and nanochemistry.
The present study was undertaken to investigate the safe doses of Pueraria tuberosa water extract (PTWE) on different organs. The OECD guidelines 407 of repeated toxicity was followed with respect to the selection of dose and days for different organs. The selected doses of PTWE were 250, 500, 1000 and 2000 mg/kg b wt for 7, 14, 21 and 28 days. Haematoxylin and eosin staining was used to study the morphological alterations in heart, intestine, testis, adrenal gland and spleen. In the present study, no adverse alterations in cardiac fibers of the heart, size and shapes in crypts and villi of intestine, seminiferous tubules and spermatozoa count in testis, three zones of adrenal gland, and spleen were seen in all treated groups of PTWE. There were no adverse morphological alterations found in described organs. The PTWE are safe at 1000 mg/kg b wt. up to 28 days and 2000 mg/ kg b. wt up to 21 days, respectively.
BACKGROUND:Pelvic lymph node (PLN) metastasis has been included in the FIGO staging, so there is a need to determine the dose contribution from brachytherapy to ascertain the total delivered dose to the pelvic lymph nodes in cervical cancer. The aim of the study was to calculate the equivalent dose in 2 Gy (EQD2) of the pelvic lymph nodes (PLNs) based on volume prescription using three applicators.MATERIALS AND METHODS:Forty-one patients who had undergone external beam radiotherapy followed by brachytherapy using tandem ovoids (TO), tandem ring (TR) and TO + free hand interstitial needles (TO + FH) applicators were taken for this study. 26 Gy in 4 fractions was prescribed to HRCTV. The external iliac node (ELN), internal iliac node (ILN) and obturator (OBT) were contoured and the median EQD2 of the lymph nodes was calculated.RESULTS:The median bilateral EQD2 values of ELN were 1.55 Gy (TR), 1.75 Gy (TO), 1.9 Gy (TO + FH), of ILN these were 2.57 Gy (TR), 3.27 Gy (TO), 3.04 Gy (TO + FH), and of OBT these were 3.69 Gy (TR), 4.46 Gy (TO), 4.69 Gy (TO + FH), respectively. The total median EQD2 values of TR, TO and TO + FH were 52.71 Gy, 53.03 Gy, and 53.88-62.73 Gy, respectively.CONCLUSION:Our study calculated the median EQD2 to the pelvic lymph nodes using three types of applicators in brachytherapy. This could serve as reference to decide on the EBRT boost dose while treating patients with enlarged pelvic lymph nodes.
Demand and conservation for potable water has become a foremost concern world-wide. Many technologies were adapted for converting the saline water to potable water to meet the required demand on water conservation. In the current research work triangular solar still with rectangular-fins attached to the basin is proposed to enhance the output of potable water from the solar still setup. Solar still with and without rectangular-fins on the basin are fabricated for experimental comparison and evaluation in addition to numerical investigations. Thermal Performance, instantaneous efficiency and potable water output of the proposed solar still & base solar still are investigated during March month for the location of Sathyamangalam. Investigation shows enhancement of water production in the proposed solar still by 41% higher compared to the base still. The maximum distillate output from modified still and base still for a typical day is 3.1 liter and 2.2 liter respectively.
Expanding and refining the skill sets of postgraduates specialists is required by the many current and changing demands of dental science education and research. To meet this challenge, the Indian Board of Micro Restorative and Endodontics under the aegis of the Indian Association of Conservative Dentistry and Endodontics has begun a fellowship program in Micro Restorative and Endodontics; the main aim being to improve the knowledge and skills of these specialists empowering them to go a step higher and beyond their areas of specialization. The objectives of the fellowship course are as follows: (1) Achieving necessary skills in managing routine and advanced restorative and endodontic scenarios using modern magnification technology. (2) Constantly enhancing the existing knowledge by pursuing new arenas in magnification technology. (3) Complete understanding and application of cutting edge research in the practice of microrestorative and endodontics. (4) Ensuring the highest possible patient-centric quality of care. Learning experiences shall be provided under the guidance and supervision of competent mentors at predesignated centers all over India. The learning strategies will be through workshops, contact sessions, and learning assignments. The carefully designed curriculum shall prepare the restorative dentists and endodontists, to be an autonomous, effective, safe, and a compassionate professional, who practices collaboratively in a variety of clinical and academic setups, responsive to the current and future needs of the restorative and endodontic health-care system. A passion for microdentistry would be a great way to make a positive difference to your patient care and creative work.
Context: This study was carried out to assess the currently available composite systems in India for resistance to fracture in teeth with large cavities. Aim: To evaluate the fracture toughness of weakened maxillary premolars restored by contemporary composites. Settings and Design: In-vitro study was done in the department of conservative dentistry and endodontics. Materials and Methods: Freshly extracted 44 human maxillary bicuspids were randomly divided into four groups where Group I included intact teeth. The teeth in the other three groups were subjected to a standardized mesio-occluso-distal cavity preparation. The cavities were filled in Group II with bioactive, nanohybrid composite with Giomer chemistry (Beautifil II LS, Shofu Inc., Kyoto, Japan), in Group III with highly filled, nanohybrid composite (Prevest Fusion Universal, Prevest DenPro Limited, Jammu, India) and in Group IV with fiber-reinforced composite with optimized fiber-aspect ratio technology (everX Posterior, GC EUROPE N. V., Leuven, Belgium). All the specimens were then subjected to thermocycling followed by incubation procedures. Fracture resistance was measured in Newton (N) using the universal testing machine. Statistical Analysis Used: Data obtained were tabulated and subjected to one-way analysis of variance (ANOVA) test followed by Tukey's test. Results: Highest mean fracture resistance was observed with Group I (2294.06), followed by Group II (1708.72), Group IV (1195.82), and Group III (825.38). One-way ANOVA test showed a statistically significant difference (P ≤ 0.05) between all the four groups. Post hoc Tukey test was used for intergroup comparison and showed significant difference (P ≤ 0.05) in mean fracture resistance between groups. Conclusion: The results suggest that the highest compressive fracture resistance was shown by Group II (Beautifil II LS composite resin).
Functionally important, local structural transitions in DNA generate various alternative conformations. Cruciform is one of such alternative DNA structures, usually targeted in genomes by various proteins. Symmetry elements in sequence as inverted repeats are the key factor for cruciform formation, facilitated by the presence of the AT-rich regions. Here, we used biophysical and biochemical techniques such as Gel electrophoresis, Circular dichroism (CD), and UV-thermal melting analysis to explore the structural status of the designed DNA sequences, which had potential to form cruciform structures under physiological conditions. The gel electrophoresis analysis revealed that the designed 53-mer DNA oligonucleotide sequence CR forms an intermolecular bulge duplex with flanking ends, while another sequence CRC adopts an intramolecular hairpin structure with flanking ends. Their equimolar complex (CRCRC) bestowed much-retarded migration due to the formation of a quite intriguing cruciform structure. CD studies confirmed that all the alternative structures (cruciform, bulge duplex, and hairpin with flanking ends) exhibit characteristics of B-DNA type conformation. A triphasic UV-thermal melting curve displayed by the complex formed by the equimolar ratio (CRCRC) is also suggestive of the formation of the cruciform structure. The interaction studies of CR, CRC, and their equimolar complex (1:1) with a photosensitizer methylene blue (MB) indicated that MB could not stabilize the discrete structures formed by CR and CRC sequences, however, the cruciform structure showed a quite significant increment in the melting temperature. Such studies facilitate our understanding of various secondary structures possibly present inside the cell and their interactions with drug/dye molecules.
The NCBI database has > 15 chloroplast (cp) genome sequences available for different Camellia species but none for C. assamica. There is no report of any mitochondrial (mt) genome in the Camellia genus or Theaceae family. With the strong believes that these organelle genomes can play a great tool for taxonomic and phylogenetic analysis, we successfully assembled and analyzed cp and mt genome of C. assamica. We assembled the complete mt genome of C. assamica in a single circular contig of 707,441 bp length comprising of a total of 66 annotated genes, including 35 protein-coding genes, 29 tRNAs and two rRNAs. The first ever cp genome of C. assamica resulted in a circular contig of 157,353 bp length with a typical quadripartite structure. Phylogenetic analysis based on these organelle genomes showed that C. assamica was closely related to C. sinensis and C. leptophylla. It also supports Caryophyllales as Superasterids.
The mechanism of fat soluble bioactives on activation of AMPK, normalization of insulin and serum proteome on obesity is not yet clear. This study investigated the effect of wheat bran oil concentrate on adipokines, pro-inflammatory markers and transcription factors in relation to differentially expressed serum proteins using two-dimensional electrophoresis (2DE) approach-in high fat diet (HFC) fed C57BL6 mice.The HFC group exhibited a significant increase in body weight, insulin level and lipid profile. The ELISA and Western blot analysis of serum proteins of HFC group also exhibited increased level of inflammatory markers (TNF-α, TGF-β and interleukin-1β), tissue transcription factors (SREBP-2 & PPARγ) and were validated by gene expression and immunohistochemistry. However, the treatment with WBA 1 and 2% significantly lessened lipid droplets in 3T3-L1 cells. Reduced insulin level, down-regulated transcription factors and associated inflammatory cytokines by activating AMPK in HFC-fed mice. Furthermore, WBA treatment directly reversed differentially expressed recombinant proteins of KCTD family and Zinc finger protein 236 isoform.Bioactive fraction of WBA had beneficial effect on ameliorating obesity related complications mainly through phosphorylation of AMPK and inflammatory cytokines which was authenticated by proteomic analysis.
Abstract Tea is a highly cross-pollinated, woody, perennial tree. High heterozygosity combined with a long gestational period makes conventional breeding a cumbersome process. Therefore, marker-assisted breeding is a better alternative approach when compared with conventional breeding. Considering the large genome size of tea (~3 Gb), information about simple sequence repeat (SSR) is scanty. Thus, we have taken advantage of the recently published tea genomes to identify large numbers of SSR markers in the tea. Besides the genomic sequences, we identified SSRs from the other publicly available sequences such as RNA-seq, GSS, ESTs and organelle genomes (chloroplasts and mitochondrial) and also searched published literature to catalog validated set of tea SSR markers. The complete exercise yielded a total of 935 547 SSRs. Out of the total, 82 SSRs were selected for validation among a diverse set of tea genotypes. Six primers (each with four to six alleles, an average of five alleles per locus) out of the total 27 polymorphic primers were used for a diversity analysis in 36 tea genotypes with mean polymorphic information content of 0.61–0.76. Finally, using all the information generated in this study, we have developed a user-friendly database (TeaMiD; http://indianteagenome.in:8080/teamid/) that hosts SSR from all the six resources including three nuclear genomes of tea and transcriptome sequences of 17 Camellia wild species. Camellia assamica L. (O). Kuntze. var. TV-1 from Assam, India is one of the most important tea cultivar of Indian Tea Industry which botanically belongs to Assam type and has distinct morphological as well as chemical constituent in leaf than China type of tea. We here present the first draft genome sequence of TV-1, assembled in 2.93 Gb size comprising of 14,824 scaffolds and covering 97.66% of the predicted genome size (3.0 Gb) as determined by flow cytometry. The N50 value, longest and average contig size for this draft genome was found to be as 538.96 Kb, 3.64 Mb and 197.99 Kb, respectively. There were 495,747 SSRs (Di- to Hexa-mers) in the genome and the repeat contents accounted for 71.87% of the genome. About 94.40% genome completeness was predicted with BUSCO, maximum among the published and reported tea genomes. The clustering of the assembled genome with Hi-C data resulted in 15 clusters ranging from the size of 631.46 Mb to 14.39 Mb and accounting 99.32% of assembled genome. This data will act as the starting point for unravelling the important genes and differentiating Indian tea from other commercially available tea varieties.
