Tomato (Solanum lycopersicum L.) productivity is often limited by biotic stresses and the use of lowperforming cultivars, necessitating the development of superior hybrids. The present study was conducted during at SKUAST-Jammu, Chatha, under protected cultivation to assess heterosis and combining ability for yield and related traits in tomato. Fifteen lines and three testers were crossed using a Line × Tester design to produce 45 F1 hybrids, which were evaluated along with parents and a commercial check under low-cost polyhouse conditions. Heterotic vigor was present for growth and yield characters among hybrids. Significant genetic variability was observed for all traits studied. Several hybrids exhibited desirable heterosis for fruit yield, fruit number, fruit size, and quality attributes. Negative heterosis for days to 50% flowering indicated the potential for developing early maturing hybrids. Hybrids such as Arka Abha × EC 521038, Roma × EC 521038, and Punjab Varkha Bahar-4 × EC 521038 performed well for yield, whereas Pusa Ruby × EC 521038 and CLN-2123-A1 Red × Punjab Sartaj were superior for quality traits. Heterosis breeding is used to improve yield quantity and quality of Tomato (Solanum lycopersicum L.) because traditional methods cannot be used to achieve this goal. The study demonstrates the usefulness of heterosis breeding for developing high-yielding tomato hybrids under protected conditions
Introduction Peripheral pulse oximeters with continuous monitoring of heart rate and saturation have revolutionized neonatal care. Motion artifacts, hypothermia, and poor perfusion are some of the barriers for reliable reading in the transmittance type of peripheral pulse oximeters. This study was planned to assess the effectiveness of transmittance sensors applied over the forehead for monitoring the heart rate and saturation. Material and Methods An observation study was conducted over a period of 1 month. Two pulse oximeters (Masimo RAD97) were applied simultaneously, one over the periphery (right hand/wrist) and the other over the forehead using an innovative headband. A total of 540 readings of heart rate and saturation (SpO2) from each site were recorded. A difference of more than 5 beats/minute in heart rate and 2% saturation were considered clinically significant. Results Forty-five neonates with mean gestational age of 35.3 ± 3.2 weeks and birth weight of 2109 ± 683 grams were enrolled. Forehead pulse oximeter could pick up the heart rate and SpO2 readings in all the babies. A statistically significant difference of 3.8 beats/minute in heart rate and 2.5% in SpO2 was noted ( p-value < 0.0001). The difference in heart rate was not clinically significant. Conclusion We propose that the transmittance type peripheral pulse oximeter sensors can be used over the forehead. It has the potential to avoid erroneous readings due to motion artifacts, hypothermia, or shock. Saturation nomograms for the forehead pulse oximetry need to be established before it can be used to monitor and manage the neonates for the same.
Global food demand is expected to increase between 55 and 70% by 2050. Plant breeders and geneticists are constantly under pressure to develop high-yielding climate-resilient varieties using novel approaches. The quest for simplifying complex traits and efforts for developing high-yielding varieties during the twenty-first century led to a paradigm shift from phenotypic-based selection to genome-based breeding. On one hand, the development and utilization of diverse genetic resources, and advances in genomics on the other hand provided a kick start for the understanding the genetics of economically important complex traits at a faster pace. Further, the next-generation sequencing revolutionized our understanding of the genome architecture. As a result, there has been an increasing demand for statistical and bioinformatics tools to analyse and manage the enormous amount of data generated from sequencing of genomes, transcriptomes, proteome and metabolomes. In this chapter, we review the intervention of bioinformatics and computational tools for deploying the tremendous wealth of data for plant genetics and breeding research.
Dear Editor, The SARS-CoV-2 pandemic had caused widespread disruption globally, but a safe and effective vaccine was developed within a year.[1,2] India started its vaccination drive on January 16, 2021, and as of September 30, 2022, over 2 billion COVID-19 vaccines have been administered. India began COVID-19 vaccination for children aged 15–18 on January 3, 2022, and 61,933,703 doses have been administered to adolescents.[3] The leading vaccines in India are Covishield and Covaxin. It has been observed that in the initial days of the COVID-19 vaccination campaign, launched in India on January 16, 2021, there was significant vaccine hesitancy among health-care workers.[4] The hesitancy somewhat decreased in the second wave of the pandemic. However, vaccine hesitancy remains among the target population, with significant anxiety about the vaccine. Understanding the perception and attitude of parents of children below 15 years of age is crucial for designing effective communication campaigns to improve vaccine uptake. This letter highlights the findings of a study aimed at understanding vaccine hesitancy among Indian parents of children aged 1–15 years in a sub-Himalayan region and associated parental concerns associated with it. The study surveyed a group of Indian parents, focusing on their knowledge and attitudes towards the COVID-19 vaccine. Data were collected through one-to-one interviews using a structured questionnaire. The Vaccine Attitudes Examination Scale was used to measure hesitancy. The mean age of the participants was 39.94 ± 13.44 years, with 58.6% being male. The average age of their children was 7.49 ± 4.08 years. Most respondents, 127 (34.1%), had less than primary education, and 235 (63.2%) lived in nuclear families. Eighty-six (23.1%) of the respondents had previously experienced COVID-19, and 83.1% had taken the vaccine. The average perceived knowledge about the COVID-19 vaccine on a scale of 1 to 10 was 4.98 ± 1.94, with most knowing Covishield and Covaxin. The source of vaccine information was television in 166 (44.62%) respondents, followed by the Internet in 127 (34.14%) Vaccine acceptance was 72.6% if no cost was involved, which dropped to 48.7% if the vaccine incurred cost. The main concern among parents is the adverse effects of the vaccine (54.03%), followed by concerns about the vaccine's safety and efficacy. No significant association was found between parents' willingness to administer the vaccine and gender, COVID diagnosis, or adverse effects. Vaccine hesitancy arises from an interwoven multitude of factors involving demographic, psychological, and political factors; vaccine hesitancy needs to be addressed to control not only the current pandemic but also the forthcoming pandemics. On the individual level, one should address the perceived risks of taking the vaccines, and on the community level, better dissemination of the benefits and safety of taking the vaccine. Now and in the future, meeting the global demand for healthy food for the ever-increasing population is a crucial challenge. In the last seven decades, agricultural practices have shifted to the use of synthetic fertilizers and pesticides to achieve higher yields. Despite the huge contribution of synthetic fertilizers in agronomy, their adverse effects on the environment, natural microbial habitat, and human health cannot be underrated. Besides, synthetic fertilizers are manufactured from non-renewable sources such as earth mining or rock exploitation. In this context, understanding and exploiting soil microbiota appears promising to enhance crop production without jeopardizing the environment and human health. This chapter reviews the historical as well as current research efforts made in identifying the interaction between soil microbes and root exudates for crop improvement. First, microbial consortium viz. bacteria, algae, fungi, and protozoa are briefly discussed. Then, the application of bio-stimulants followed by genome editing of microbes for crop improvement is summarized. Finally, the perspectives and opportunities to produce bioenergy and bio-fertilizers are analyzed.
Objective: We aim to analyze the efficacy and safety of TMS on cognition in mild cognitive impairment (MCI), Alzheimer's disease (AD), AD-related dementias, and nondementia conditions with comorbid cognitive impairment. Design: Systematic review, Meta-Analysis Setting: We searched MEDLINE, Embase, Cochrane database, APA PsycINFO, Web of Science, and Scopus from January 1, 2000, to February 9, 2023. Participants and interventions: RCTs, open-label, and case series studies reporting cognitive outcomes following TMS intervention were included. Measurement: Cognitive and safety outcomes were measured. Cochrane Risk of Bias for RCTs and MINORS (Methodological Index for Non-Randomized Studies) criteria were used to evaluate study quality. This study was registered with PROSPERO (CRD42022326423). Results: The systematic review included 143 studies (n = 5,800 participants) worldwide, encompassing 94 RCTs, 43 open-label prospective, 3 open-label retrospective, and 3 case series. The meta-analysis included 25 RCTs in MCI and AD. Collectively, these studies provide evidence of improved global and specific cognitive measures with TMS across diagnostic groups. Only 2 studies (among 143) reported 4 adverse events of seizures: 3 were deemed TMS unrelated and another resolved with coil repositioning. Meta-analysis showed large effect sizes on global cognition (Mini-Mental State Examination (SMD = 0.80 [0.26, 1.33], p = 0.003), Montreal Cognitive Assessment (SMD = 0.85 [0.26, 1.44], p = 0.005), Alzheimer's Disease Assessment Scale-Cognitive Subscale (SMD = -0.96 [-1.32, -0.60], p < 0.001)) in MCI and AD, although with significant heterogeneity. Conclusion: The reviewed studies provide favorable evidence of improved cognition with TMS across all groups with cognitive impairment. TMS was safe and well tolerated with infrequent serious adverse events.
Carotenoids are the natural pigments available in nature and exhibit different colors such as yellow, red, and orange. These are a class of phytonutrients that have anti-cancer, anti-inflammatory, anti-oxidant, immune-modulatory, and anti-aging properties. These were used in food, pharmaceutical, nutraceutical, and cosmetic industries. They are divided into two classes: carotenes and xanthophylls. The carotenes are non-oxygenated derivatives and xanthophylls are oxygenated derivatives. The major source of carotenoids are vegetables, fruits, and tissues. Carotenoids also perform the roles of photoprotection and photosynthesis. In addition to the roles mentioned above, they are also involved and act as precursor molecules for the biosynthesis of phytohormones such as strigolactone and abscisic acid. This chapter briefly introduces carotenoids and their extraction method from plant tissue. Proposed protocol describes the extraction of carotenoid using solvents chloroform and dichloromethane. Reverse-phase HPLC can be performed with C30 columns using gradient elution. The column C30 is preferred to the C18 column because the C30 column has salient features, which include selective nature in the separation of structural isomers and hydrophobic, long-chain compounds, and shows the best compatibility with highly aqueous mobile phases. A complete pipeline for the extraction of carotenoids from plant tissue is given in the present protocol.
Pre-harvest sprouting (PHS) in groundnut leads to substantial yield losses and reduced seed quality, resulting in reduced market value of groundnuts. Breeding cultivars with 14–21 days of fresh seed dormancy (FSD) holds promise for precisely mitigating the yield and quality deterioration. In view of this, six multi-locus genome-wide association study (ML-GWAS) models alongside a single-locus GWAS (SL-GWAS) model were employed on a groundnut mini-core collection using multi season phenotyping and 58 K “Axiom_Arachis” array genotyping data. A total of 9 significant SNP-trait associations (STAs) for FSD were detected on A01, A04, A08, A09, B02, B04, B05, B07 and B09 chromosomes using six ML-GWAS models. Additionally, the SL-GWAS model identified 38 STAs across 14 chromosomes of groundnut. A single STA on chromosome B02 (qFSD-B02-1) was consistently identified in both ML-GWAS and SL-GWAS models. Furthermore, candidate gene mining identified nine high confidence genes viz., Cytochrome P450 705 A, Dormancy/auxin associated family protein, WRKY family transcription factor, Protein kinase superfamily protein, serine/threonine protein phosphatase, myb transcription factor, transcriptional regulator STERILE APETALA-like, ethylene-responsive transcription factor 7-like and F-box protein interaction domain protein as prime regulators involved in Abscisic acid/Gibberellic acid signaling pathways regulating dormancy/germination. In addition, three of the allele-specific markers developed from the identified STAs were validated across a diverse panel. These markers hold potential for increasing dormancy in groundnut through marker-assisted selection (MAS). Thus, this research offers insights into genetic and molecular mechanisms underlying groundnut seed dormancy in addition to providing markers and donors for breeding future varieties with 2–3 weeks of FSD.

Plant metabolomics research aims to detect precisely all metabolites found within tissues of plants through GC-MS. This chapter primarily focuses on extracting metabolites using chemicals such as methanol, chloroform, ribitol, MSTFA, and TMCS. The metabolic analysis method is frequently used according to the specific kind of sample or matrix being investigated and the analysis objective. Chromatography (LC, GC, and CE) with mass spectrometry and NMR spectroscopy is used in modern metabolomics to analyze metabolites from plant samples. The most frequently used method for metabolites analysis is the GC-MS. It is a powerful technique that combines gas chromatography's separation capabilities with mass spectrometry, offering detailed information, including structural identification of each metabolite. This chapter contains an easy-to-follow guide to extract plant-based metabolites. The current protocol provides all the information needed for extracting metabolites from a plant, precautions, and troubleshooting.
BackgroundWith ever increasing rates of emergency caesarean deliveries (CD),incorporating the ERAS protocol might provide a perfect window of opportunity to increase maternal comfort during the postsurgical period, but also improve outcomes and facilitate optimal return of physiological function.ObjectiveTo determine whether an ERAS pathway at emergency caesarean birth would permit a reduction in postoperative length of stay and improve postoperative patient satisfaction.Material & MethodsPatients undergoing emergent caesarean delivery at ≥ 34 weeks of gestation were randomized to ERAS or conventional care. The primary outcome was to compare postoperative length of hospital stay. Secondary outcome variables included first oral intake, passage of flatus/defecation, first ambulation, first urination after catheter removal and postoperative pain scores in both groups.ResultsWe randomized 142 women (71 each in ERAS versus Conventional arm) undergoing emergency cesarean delivery. Incorporation of ERAS protocol resulted in shorter length of hospital stay (73.92 ± 8.96 in conventional arm vs 53.87 ± 15.02 in ERAS arm; p value <.0001). Significant difference was seen in visual analogue scoring during initial ambulation and rest on day 0 and day 1 between ERAS and conventional arms with mean scores being lower in ERAS arm compared to Conventional arm (p value <.05). In terms of quality of life, ERAS arm had better quality of life compared to conventional armConclusionIncorporation of ERAS protocol in emergency caesarean definitely improves patient outcome in terms of early resumption of activities with better quality of life.
Background Maternal COVID-19 infection acquired during late pregnancy carries a potential risk for adverse neonatal outcomes. There is still a paucity of data on its effect on the transition from intrauterine to extrauterine life. Objectives The objectives of this study were to determine the impact of maternal COVID-19 infection on neonates for the risk and need for resuscitation at birth, Apgar scores at 1- and 5-minutes, and the need of NICU admission during early neonatal period. Materials and method In this hospital-based prospective matched cohort study, 100 COVID-positive pregnant women presenting for delivery were enrolled. We also included 100 non-COVID pregnant women after the best possible matching of their major baseline parameters with the study group. Neonates of both groups were followed-up till 7 days of life. Results The two groups were comparable for all baseline variables except for the mode of delivery. The requirement of neonatal resuscitation was 30% and 21% in the study and control groups (RR = 1.429; 95% CI 0.88–2.32; p = 0.149). Apgar scores at 1- and 5- minutes were also unaffected by maternal COVID-19 infection with mean scores of 8.8 ± 0.651 vs. 8.87 ± 0.562 (p = 0.42) in the study and control groups, respectively. COVID-exposed neonates had a higher incidence of NICU admission when compared with the unexposed group (RR =1.616; 95% CI 1.002–2.606; p = 0.047). Among neonates born to COVID-positive mothers, 11% demonstrated evidence of SARS-CoV-2 positivity within first 5 days of life. The risk for need of resuscitation and mean Apgar scores were comparable among SARS-CoV-2 positive and negative neonates (p > 0.05). Conclusion COVID-19 infection in pregnant women is not associated with an increased risk of neonatal resuscitation.
Pneumopericardium (PPC) is a rare but life-threatening condition that mostly accompanies air leak syndromes in neonates receiving positive pressure ventilation. PPC results from the rupture of the alveoli and subsequent dissection of air through the tissue planes into the mediastinum, pleura, or pericardial sac. This report describes the case of a term low birth weight neonate born in a peripheral hospital who developed respiratory distress soon after birth. She was transported with manual bag and tube ventilation, and was found to have air leak syndrome in the form of isolated PPC. The neonate was managed conservatively with spontaneous resolution of the PPC.
Lithium has been a cornerstone treatment for bipolar disorder (BD). Despite descriptions in the literature regarding associations between long-term lithium therapy (LTLT) and development of a thyroid disorder (overt/subclinical hypo/hyperthyroidism, thyroid nodule, and goiter) in BD, factors such as time to onset of thyroid abnormalities and impact on clinical outcomes in the course of illness have not been fully characterized. In this study we aimed to compare clinical characteristics of adult BD patients with and without thyroid disorders who were on LTLT. We aimed to identify the incidence of thyroid disorders in patients with BD on LTLT and response to lithium between patients with and without thyroid disorders in BD. The Cox proportional model was used to find the median time to the development of a thyroid disorder. Our results showed that up to 32% of patients with BD on LTLT developed a thyroid disorder, of which 79% developed hypothyroidism, which was corrected with thyroid hormone replacement. We did not find significant differences in lithium response between patients with or without thyroid disorders in BD. Findings from this study suggest that patients with BD and comorbid thyroid disorders when adequately treated have a response to lithium similar to patients with BD and no thyroid disorders.
Context:Hypospadias is a common urological anomaly which could be surgically corrected with good cosmetic results.Aims:We aimed to detect changes in urinary flow parameters both before and after tubularised incised plate urethroplasty (TIPU) using uroflowmetry.Settings and Design:Data collected were clinically implemented hypothesising the probability of urethrocutaneous fistula following stricture with Qmax variation.Materials and Methods:This study is a prospective analysis done from December 2017 to October 2019. A total of 104 cases of anterior hypospadias were included in the study. A single surgical unit did TIPU. Pre-operative and post-operative uroflowmetry was done, and Qmax was recorded at 3 months, 6 months and 1 year after surgery. Mean Qmax was calculated for all intervals. A significant decrease in Qmax of a child (<2 standard deviation) was ascertained. Urethral calibration was done in those cases with a significant decrease of Qmax and analysed statistically.Results:The mean age was 6.97 ± 2.41 years. Out of 104 children, 73 (70.2%) and 31 (29.8%) had distal and mid-shaft hypospadias, respectively. The pre-operative mean Qmax of the population was 6.20 ± 0.42 ml/s. Arithmetic mean Qmax at 3 months, 6 months and 1 year was 8.53 ± 0.42, 11.18 ± 0.47 and 13.71 ± 0.44 ml/s, respectively. On comparing the pre-operative with post-operative mean Qmax, a significant increase was found postoperatively (P < 0.0001). Twenty-four patients had significantly decreased Qmax value after 6 months. In these patients, follow-up urethral dilation was done with significant improvement.Conclusion:The changes in maximum flow rate (Qmax) are suitable for use in routine follow-up. A significant decrease in Qmax over time indicates the onset of urethral stricture. These cases are to be intervened before venturing to redo urethroplasty.
Objective: To investigate the association between thyroid-stimulating hormone (TSH) and clinically relevant depression (CRD) in a population-based study. Patients and Methods: Adult patients (>18 years of age) who received care at Mayo Clinic in Rochester, Minnesota, and completed a TSH and Patient Health Questionnaire -9 (PHQ-9) within 6 months of each other, between July 8, 2017, and August 31, 2021, were included. Demographics, medical comorbidities, thyroid function laboratory data, psychotropic medications, presence of pri-mary thyroid disorder, thyroid hormone replacement (T4 and/or T3), and mood disorder diagnoses (using International Classification of Diseases, 10th version, Clinical Modifications codes) were extracted electronically. The primary outcome, CRD, was defined as a PHQ-9 score greater than or equal to 10. Logistic regression analysis was conducted to assess the association between TSH cate-gories (low <0.3 mIU/L; normal >0.3-4.2 mIU/L; high >4.2 mIU/L) and CRD. Results: The cohort included 29,034 patients, mean age 51.4 years, 65% females, 89.9% White, and a mean body mass index of 29.9 kg/m2. The mean & PLUSMN; standard deviation for TSH was 3.0 & PLUSMN;8.5 mIU/L, and the mean PHQ-9 score was 6.3 & PLUSMN;6.2. After adjustment, the odds of CRD were significantly higher among the low TSH category (odds ratio, 1.37; 95% CI, 1.18-1.57; P<.001) compared with the normal TSH category, especially in people 70 years of age or younger compared with people older than 70 years of age. Subgroup analysis did not show an increase in odds of CRD among patients with sub-clinical/overt hypothyroidism/hyperthyroidism (after adjustment). Conclusion: In this large population-based cross-sectional study, we report that low TSH was asso-ciated with higher odds of depression. Future longitudinal cohort studies are needed to investigate the relationship between thyroid dysfunction and depression as well as sex differences. & COPY; 2023 Mayo Foundation for Medical Education and Research. In addition to the challenge of meeting global demand for food production, there are increasing concerns about food safety and the need to protect consumer health from the negative effects of foodborne allergies. Certain bio-molecules (usually proteins) present in food can act as allergens that trigger unusual immunological reactions, with potentially life-threatening consequences. The relentless working lifestyles of the modern era often incorporate poor eating habits that include readymade prepackaged and processed foods, which contain additives such as peanuts, tree nuts, wheat, and soy-based products, rather than traditional home cooking. Of the predominant allergenic foods (soybean, wheat, fish, peanut, shellfish, tree nuts, eggs, and milk), peanuts (Arachis hypogaea) are the best characterized source of allergens, followed by tree nuts (Juglans regia, Prunus amygdalus, Corylus avellana, Carya illinoinensis, Anacardium occidentale, Pistacia vera, Bertholletia excels), wheat (Triticum aestivum), soybeans (Glycine max), and kidney beans (Phaseolus vulgaris). The prevalence of food allergies has risen significantly in recent years including chance of accidental exposure to such foods. In contrast, the standards of detection, diagnosis, and cure have not kept pace and unfortunately are often suboptimal. In this review, we mainly focus on the prevalence of allergies associated with peanut, tree nuts, wheat, soybean, and kidney bean, highlighting their physiological properties and functions as well as considering research directions for tailoring allergen gene expression. In particular, we discuss how recent advances in molecular breeding, genetic engineering, and genome editing can be used to develop potential low allergen food crops that protect consumer health.
Abstract Purpose Long-term lithium therapy (LTLT) has been associated with chronic kidney disease (CKD). We investigated changes in clinical characteristics, pharmacotherapeutic treatments for medical/psychiatric disorders, and outcomes among patients with bipolar disorder (BD) and CKD on LTLT in a 2-year mirror-image study design. Methods Adult BD patients on LTLT for ≥1 year who enrolled in the Mayo Clinic Bipolar Disorder Biobank and developed CKD (stage 3) were included, and our study was approved by the Mayo Clinic Institutional Review Board. The primary outcome was the time to the first mood episode after CKD diagnosis among the lithium (Li) continuers and discontinuers. Cox proportional hazards models were used to estimate the time to the first mood episode. We tested for differences in other medication changes between the Li continuers and discontinuers group using Mantel-Haenszel χ2 tests (linear associations). Results Of 38 BD patients who developed CKD, 18 (47%) discontinued Li, and the remainder continued (n = 20). The median age of the cohort was 56 years (interquartile range [IQR], 48–67 years), 63.2% were female, and 97.4% were White. As compared with continuers, discontinuers had more psychotropic medication trials (6 [IQR, 4–6] vs 3 [IQR, 2–5], P = 0.02), a higher rate of 1 or more mood episodes (61% vs 10%, P = 0.002), and a higher risk of a mood episode after CKD diagnoses (Hazard Ratio, 8.38; 95% confidence interval, 1.85–38.0 [log-rank P = 0.001]]. Conclusions Bipolar disorder patients on LTLT who discontinued Li had a higher risk for relapse and a shorter time to the first mood episode, suggesting a need for more thorough discussion before Li discontinuation after the CKD diagnosis.
Background Mass transport of neonates is required in cases of disasters and calamities such as fire. It may also be required when there is a need for the upgradation of infrastructure. Neonatal transport even for a short period is a period of stress for the neonate. Mass transport of neonates needs much planning, and even after diligent planning, may result in the destabilization of neonates. Objectives The objectives of this study were to assess the impact of mass intrahospital neonatal transport on the vital parameters of neonates, the occurrence of any adverse event during transport. To study the frequency of adverse events in mass intrahospital neonatal transport and factors related to it. Materials and methods This was a retrospective observational study on a cohort of 16 neonates who were transported to an alternate site in the hospital so that renovation and upscaling of the infrastructure of the newborn intensive care unit (NICU) could be undertaken. Site selection, preparation, and transport details were observed. Vital parameters pre and post-transport were noted, and the occurrence of adverse events during transport was documented and analyzed. Results Sixteen neonates were transported over a span of 90 minutes with a mean travel time of 5.62±3.03 minutes. There was a statistically significant rise in the heart rate of the neonates post transport (137.7±8.51vs 141.3±9.01, p-value .00769) though not clinically significant. Six point two-five percent (6.25%) of neonates deteriorated post transport and needed extra efforts for stabilization. Equipment malfunction was responsible for deterioration. Conclusion Unforeseen events can occur during neonatal transport. Despite adequate planning, preparation, and care during transport, it remains a period of stress for a neonate.
