采用直接法制备传代细胞染色体标本、常规Giemsa染色、显微镜下计数中期分裂相染色体数目的方法对7种鳞翅目昆虫细胞系进行了染色体分析,得出了各种细胞系染色体分析所需要的秋水仙素浓度与处理时间以及低渗浓度和处理时间的范围;7种鳞翅目细胞系染色体均表现出典型的鳞翅目昆虫传代细胞的核型特征:染色体数目众多,异倍化严重,多为弥散型的着丝粒的短杆状、颗粒状、点状或圆球状.
The Chromosome analysis of 5 Diptera insect cell lines was made by direct method,with regular Giemsa staining.Chromosomes of these cells in metaphase were counted under the microscope.The chromosome numbers and their nuclear types were analyzed.C6/36 belonged to diploid cell lines and its chromosome mode was 6.SL2 belonged to hyperdiploid cell lines and its chromosome mode range was 8~12.L-2/M delta 2-3 belonged to hyperdiploid cell lines and its chromosome mode range was 8~20.Aedes albopictus Skuse belonged to diploid cell lines and its chromosome mode was 6.NIH-SaPe-4 belonged diploid or hypodiploid cell lines and its chromosome mode was 10.Chromosome numbers in the 5 cell lines were not completely changed.
The Cordyceps militaris found in Zhaotong,Yunnan Province was investigated.The host insect is Biston falcata,which has one generation a year in Zhaotong and overwinters by pupae.The larvae eat the needles of Pinus armandii Franch,and therefore is a harmful pest of the pine from mid September to early October.Cordyceps militaris mainly lives in armand pine forest with the altitude of 2 200~2 400 m.Its fruit bodies occur from late July to late September,in the mild,moist and rainy habitat.Analysis indicated that the fungus had rich nutrition ingredients including 646.0 g·kg-1 protein,37.0 g·kg-1 crude fat,54.3 g·kg-1 total sugar,35 g·kg-1 ash,and 17 kinds of amino acids.Total quantity of amino acids is 266.3 g·kg-1.Many kinds of inorganic elements needed in human metabolism are included.
This paper dealt with the anti-mutation experiment of white wax scale and analysis of main functional factors.The micro-nucleoli rates in every experimental dose groups were lower then the contrast in 48 h.The difference between high dose group and contrast was obvious.The restriction rate of micro-nucleoli in high dose group was 34.19%.The results showed that white wax scale could restrict mutation caused by C.P and had clear anti-mutation function.White wax scale contained 2.14% chitin.White wax scale contained 7.2% crude polysaccharide.The polysaccharide content was 4.54% after removing protein by enzyme.The sugar content of the polysaccharide was 22.1% as glucose,composed of glucose,galactose and fructose in molar ratio of 5∶21∶1.The polysaccharide isolating from white wax scale was polysaccharide-protein complex.This insect contained 1.3% flavonoid and several kinds of vitamins.chitin,polysaccharide,vitamin and flavonoid were main anti-mutation functional factors of white wax scale.
This paper dealed with the immunnodulation tests of white wax scale in specific and unspecific immunity.The result showed that the IgM in high dose group was clearly higher than that of the control in serolysin test.Lymphocyte transformation and propagation in mice were notable in high dose group.The results of carbon clearance test in mice and the weight of immune organ test(unspecific immune tests)were negative.The research results showed that white wax scale could increase the specific humoral and cell immunity ability. Therefore white wax scale has immunnodulation function which could be developed and utilized as health food resource.
