Luís P. Fonseca is an Associate Professor at the Department of Bioengineering of Instituto Superior Técnico (I.S.T.) the University of Lisbon, and Coordinator of the Biocatalysis and Biotransformation Research Group (2BRG) based in the Institute for Bioengineering and Biosciences (iBB) at I.S.T of University of Lisbon, Portugal.
After graduating in Chemical Engineering in 1983, Luis P: Fonseca got an M.Sc. in Biochemical Engineering in 1989, a Ph.D. in Biotechnology in 1995, and later his D.Sc. in Biotechnology 2006, all degrees at I.S.T.
Specific areas of research interests and expertise of Luis P. Fonseca include:
- Biosynthesis of high-value bioproducts by fermentation and implementing key enabling technologies.
- Enzyme technology and biocatalysis.
- Green chemistry based on clean environmental processes.
- Development of biomaterials and nano-composites and nano-carriers.
- High-throughput analysis for monitoring bioprocesses and pathogens.
- Synthesis of polyesters and derivatives from bio-based monomers.
- Degradation of polymers, particularly polyesters and derivatives contributing to a circular economy.
Luis P. Fonseca has developed research in Enzyme Technology and Biochemical Engineering, namely in Fermentation, downstream, and monitoring processes and, more recently, on Drug Delivery Systems.
The Fermentation processes focused on the cell growth of wild and mutant strains (e.g., E. coli, S. cerevisiae, Trichoderma reesei, and other fungi) on optimizing bioreactor performance and biosynthesis of biological products (e.g., enzymes). In this field, Luis P. Fonseca recently worked on designing and validating Microbial Fuel Cells for wastewater treatment and bioelectricity generation.
In the downstream processes, Luis P. Fonseca developed integrative processes of biosynthesis and in-situ recovery of enzymes using expanded bed adsorption, Aqueous Two Phase Systems, and adsorbents for pseudo-affinity chromatography.
In the monitoring processes, Luis P. Fonseca participated in the design and stabilization of biocatalysts preparations for analytical applications using immobilized multi-enzyme systems, particularly of oxidoreductases assisted by FIA/SIA systems for monitoring nutrients and metabolites (e.g., glucose, galactose, ethanol, lactate, others) in the cell culture medium. Free and immobilized proteases were tested on proteomic analysis, and metal ion sensing assisted with ultrasounds. Magnetoresistive biosensors based on microfluidic systems were also tested and developed for pathogens detection and quantification in water (e.g., E. coli, Salmonella, others). Developing a new integrative platform to discover new classes of antibiotics focusing on resistant Gram-negative bacteria, based on high-throughput FTIR spectroscopy enabling the rapid identification of the drug mechanism of action and biologic target and constitutes the core of the ASPIR Technology.
Luis P. Fonseca has a long experience developing new biocatalysis and chemo-enzymatic processes, mainly using immobilized enzymes. In Enzyme Technology, Luis P. Fonseca focused the research on material synthesis with applications ranging from semi-synthetic antibiotics, APIs, carbon-carbon bound compounds, aroma esters, biofuels, and more recently on the polymers, in particular, polyesters and derivatives.
The Biocatalytic processes have been performed in aqueous but also in non-conventional reaction mediums such as (Oil/water) miniemulsion systems that are the core of the Aromase Technology. The reversed micellar systems were also studied to solubilize enzymes in organic solvents, keeping their intrinsic enzyme activity. More recently, Luis P. Fonseca has dedicated special attention to synthesizing polyesters and derivatives in an aqueous polycondensation reaction media using immobilized enzymes and their recovery based on environmental-benign processes mainly free of organic solvents that constitute the core of the IP-protected Polymers-5B Technology.
The development of drug delivery systems for the entrapment of bioactive and hydrophobic compounds, antibiotics, and drugs in GRAS Liquid Lipid Nano-emulsions, Solid Lipid Nanoparticles, and Nanostructured Lipid Carriers are the core of NanoLipCar Technology for pharmaceutical, dermatological, or nutritional applications that can improve health and well-being.
The research of Luis P. Fonseca led to the publishing of over 170 papers in peer-reviewed journals, one national and PCT patent, graduated 16 Ph.D. students and supervised 6 Post-doctoral scientists, and participated in 20 national and international projects.
